Lycorine weakens tamoxifen resistance of breast cancer via abrogating HAGLR-mediated epigenetic suppression on VGLL4 by DNMT1

Summary: This study investigated the role of lycorine, a natural alkaloid, in regulating tamoxifen resistance in breast cancer. The results showed that lycorine reduced tamoxifen resistance in breast cancer cells and increased drug-induced apoptosis. Mechanistically, lycorine modulated the expression of HAGLR and VGLL4 genes, which are involved in the resistance to tamoxifen. Overall, lycorine plays a crucial role in improving the effectiveness of tamoxifen treatment for breast cancer.

Summary: Our study shows that the downregulation of LncRNA SNHG6 is correlated with poor prognosis in pancreatic cancer patients. We demonstrate that SNHG6 can sponge miR-944 and upregulate the target gene KPNA5, resulting in suppression of cancer cell proliferation and increased sensitivity to GEM treatment in GEM-resistant pancreatic cancer cells. Moreover, we find that higher expression of KPNA5 is associated with GEM non-resistance in patients. In summary, our findings suggest that targeting the SNHG6/miR944/KPNA5 axis may enhance the efficacy of GEM treatment in pancreatic cancer.

LncRNA-NEF suppressed oxaliplatin resistance and epithelial-mesenchymal transition in colorectal cancer through epigenetically inactivating MEK/ERK signaling

Summary: Acquired epithelial-mesenchymal transition is a major cause of oxaliplatin chemoresistance in colorectal cancer (CRC). In this study, it was found that lncRNA Neighboring Enhancer of FOXA2 (lncRNA-NEF) was down-regulated in an oxaliplatin-resistant CRC cell line. Further studies showed that lncRNA-NEF suppressed oxaliplatin resistance and epithelial-mesenchymal transition both in vitro and in vivo by promoting the expression of DOK1 and inhibiting the MEK/ERK signaling pathway. This research reveals the critical role of lncRNA-NEF in mediating oxaliplatin chemotherapy resistance in CRC and suggests a potential therapeutic strategy for CRC patients with oxaliplatin resistance.

Transcription factor B-MYB activates lncRNA CCAT1 and upregulates SOCS3 to promote chemoresistance in colorectal cancer

Summary: According to bioinformatic analysis and experimental verification, both long non-coding RNA CCAT1 and upstream gene B-MYB are overexpressed in colorectal cancer (CRC) cells. It was found that CCAT1 promotes the resistance of CRC cells to oxaliplatin (OXA) by activating B-MYB, which further recruits DNMT1 to inhibit the expression of SOCS3, leading to increased resistance to OXA. These findings suggest that B-MYB may promote chemoresistance of CRC cells to OXA through the CCAT1/DNMT1/SOCS3 axis.

Summary: In this study, a novel upregulated long non-coding RNA (lncRNA) called CACClnc was identified, which was associated with chemoresistance and poor prognosis in colorectal cancer (CRC). The study revealed that CACClnc promoted CRC resistance to chemotherapy by enhancing DNA repair and homologous recombination. The expression of exosomal CACClnc in peripheral plasma of CRC patients was also found to effectively predict the chemotherapy effect prior to treatment. LncSNHG14 promotes nutlin3a resistance by inhibiting ferroptosis via the miR-206/SLC7A11 axis in osteosarcoma cells

Summary: Osteosarcoma (OS) is the most common primary osseous malignant tumor in adolescents and children. The standard therapy involves surgery combined with neoadjuvant/post-surgery chemotherapy, but chemoresistance often leads to poor efficacy. In this study, the upregulation of lncRNA SNHG14 was found to contribute to treatment resistance by suppressing ferroptosis in a nutlin3a-resistant OS cell line. Knockdown of lncRNA SNHG14 reversed drug resistance and activated ferroptosis, indicating its role in chemotherapy resistance and ferroptosis inhibition. Targeting lncRNA SNHG14 may provide a new approach to overcome drug resistance and improve treatment efficacy in osteosarcoma.

Summary: This study found that intermittent fasting (IF) can reduce the toxicity of chemotherapy in breast cancer patients during adjuvant chemotherapy. Hematologic parameters showed no significant variations, but the glucose and insulin levels increased in the non-fasting group. The IF group showed a decrease in insulin levels. Therefore, IF can improve patients' metabolic profiles and may have a positive impact on the clinical efficacy of chemotherapy.
