Generation of stress fibers through myosin-driven reorganization of the actin cortex

Jaakko Lehtimaki et al.

Summary: The study reveals that stress fibers can form without pre-existing actomyosin bundles, giving rise to structures known as cortical stress fibers, which are embedded in the cell cortex and assemble preferentially underneath the nucleus. The assembly process is orchestrated by non-muscle myosin II pulses, leading to the reinforcement of nascent focal adhesions and subsequent stabilization of the cortical stress fibers. These findings demonstrate a new mechanism for de novo generation of stress fibers from the actin cortex, highlighting the role of stochastic myosin pulses in the assembly of functional actomyosin bundles.

Extent of myosin penetration within the actin cortex regulates cell surface mechanics

Binh An Truong Quang et al.

Summary: The study reveals that myosin does not always overlap with actin at the cortex in cells with low cortex tension, suggesting a restricted penetration. Myosin activity and actin network architecture are identified as key regulators of myosin penetration into the cortex. The spatial coordination of myosin and actin at the cortex regulates cell surface mechanics, with myosin size controlling its action by limiting minifilament penetration into the cortical actin network.

YAP-TEAD1 control of cytoskeleton dynamics and intracellular tension guides human pluripotent stem cell mesoderm specification

Stefania Pagliari et al.

Summary: The YAP-TEAD complex plays a crucial role in cellular processes such as migration, division, and differentiation. PSCs exhibit sustained YAP-driven transcriptional activity despite growing in dense colonies, which can be restored by tampering with the Tankyrase enzyme.
