Mutations in PLCZ1 induce male infertility associated with polyspermy and fertilization failure

Bi-allelic variants in ASTL cause abnormal fertilization or oocyte maturation defects

Summary: Fertilization is a critical process in development, and the inhibition of additional sperm binding after fertilization is achieved through mechanisms acting at the zona pellucida and egg plasma membrane. This study identifies bi-allelic variants in the ASTL gene, which are associated with fertilization problems in humans. The identified variants in ASTL significantly decrease the protein levels or affect the enzymatic activity involved in preventing polyspermy. Knock-in mice carrying these variants show subfertility and reduced embryo developmental potential. These findings highlight the role of ASTL in female infertility and provide a new genetic marker for diagnosing fertilization problems.

ASTL is mutated in female infertility

Summary: This study identified a consanguineous family in which adult females homozygous for a truncating variant in ASTL displayed significantly reduced fertility, similar to findings in mouse models. ASTL encodes ovastacin, which triggers zona pellucida hardening, and pathogenic variants in this gene can lead to infertility in both humans and mice. This suggests that inclusion of ASTL in female infertility gene panels may be warranted.

IQCN disruption causes fertilization failure and male infertility due to manchette assembly defect

Summary: This study used whole-exome sequencing to identify the genetic causes of total fertilization failure (TFF) in male infertility. Homozygous variants in the novel gene IQCN were found in two individuals with abnormal acrosome structures. Knockout mice lacking the Iqcn gene showed similar phenotypes to humans with TFF. The study also revealed the role of IQCN in regulating microtubule nucleation during manchette assembly, and provided a genetic marker and therapeutic option for male TFF.

Homozygous pathogenic variants in ACTL9 cause fertilization failure and male infertility in humans and mice

Summary: Genetic variations in the ACTL9 gene in male individuals have been found to cause total fertilization failure (TFF). These variations disrupt the perinuclear theca structure and affect the localization of PLCz in sperm, leading to abnormal calcium oscillations in oocytes and subsequent TFF. Overcoming TFF can be achieved through assisted oocyte activation with calcium ionophore exposure.
