Engineered extracellular vesicles for delivery of siRNA promoting targeted repair of traumatic spinal cord injury

The combination of targeted modified EVs and siRNA enhanced the anti-inflammatory and neuroreparative functions of EVs, promoting functional recovery in mice after SCI. This study highlights the potential of engineered EVs as a feasible and efficacious treatment for SCI and possibly other diseases.

Intranasal administration has been identified as an alternative approach to bypass the blood-brain barrier. In this research, polymeric micelles were prepared and characterized for the delivery of pVGF to the brain. The results showed that the modified chitosan polymer with mannose, Tat peptide, and oleic acid led to significantly higher expression of pVGF in primary astrocytes and neurons compared to intravenous administration. Intranasal administration also resulted in significantly higher pVGF expression compared to intravenous administration. These findings suggest that multifunctional polymeric micelles, especially those tagged with mannose and Tat ligands, are effective platforms for delivering pVGF to the brain.

There is a binding relationship between miR-125a and IRF5, with the knockdown of miR-125a in B-Exo leading to increased IRF5 expression and attenuation of the neuroprotective effect against SCI.
