Genome-wide Association Study Shows That Executive Functioning Is Influenced by GABAergic Processes and Is a Neurocognitive Genetic Correlate of Psychiatric Disorders

Alexander S. Hatoum et al.

Summary: This study found that deficits in executive functions are associated with psychopathology and neurologic disorders, and there is a genetic correlation with measures of intelligence and cognitive processing speed. The results suggest that executive functions are genetically distinct cognitive constructs that are particularly relevant to understanding genetic variance in psychiatric disorders.

KEGG for taxonomy-based analysis of pathways and genomes

Minoru Kanehisa et al.

Summary: KEGG is a manually curated database resource that integrates various biological objects classified into systems, genomic, chemical, and health information. Through the KEGG identifier and URL links, users can retrieve and browse information for different biological objects, including pathway maps, hierarchy, and genomes. KEGG has also improved KO assignment annotation and included more eukaryotic genomes for better representation of organisms in the taxonomic tree. The taxonomy mapping enables analysis of the conservation of functional and physical links among genes in different organism groups.

Imbalanced autophagy causes synaptic deficits in a human model for neurodevelopmental disorders

Impact of IL-21-associated peripheral and brain crosstalk on the Alzheimer's disease neuropathology

Sudhanshu Agrawal et al.

Summary: This study reveals the role of IL-21 in Alzheimer's disease (AD) pathogenesis. IL-21 enhances peripheral and brain immune and inflammatory responses, leading to increased deposition of amyloid beta (Aβ) plaques in AD. The expression of IL-21 receptor (IL-21R) is elevated in both peripheral and brain tissues of AD patients and mice.

Whole genome analysis in APOE4 homozygotes identifies the DAB1-RELN pathway in Alzheimer's disease pathogenesis

Matthew Bracher-Smith et al.

Summary: The study suggests that APOE-epsilon 4 carriers may have other genetic factors influencing the progression of Alzheimer's disease, with the discovery of a novel genome-wide significant locus associated to AD in the DAB1 gene. The analysis also indicates a potential epistatic interaction between the APOE locus and the DAB1-RELN pathway in AD.

Within-subject reaction time variability: Role of cortical networks and underlying neurophysiological mechanisms

Sivylla E. Paraskevopoulou et al.

Summary: Variations in reaction time have long been studied in human behavior, but the neural basis for within-subject and within-task variability has been minimally explored. This study used direct cortical recordings to investigate the variance in behavioral reaction time, showing that population-level neural activity differences in the task-related cortical network can explain a significant portion of the variability in reaction time. Specific brain region latencies and cumulative latency differences across the cortical network were found to contribute to reaction time variability.

APOE genotype moderates the relationship between LRP1 polymorphism and cognition across the Alzheimer's disease spectrum via disturbing default mode network

Feifei Zang et al.

Summary: This study reveals the moderating effect of APOE genotype on the relationship between LRP1 variant and DMN and cognition in AD spectrum populations. The LRP1 variant affects cognitive performance in AD spectrum populations, with altered connectivity in several brain regions observed in LRP1-T carriers. The functional connectivity of multiple brain regions within LRP1-T carriers shows strong potential for classifying AD spectrum.
