Cooperative Bimetallic Catalysis via One-Metal/Two-Ligands: Mechanistic Insights of Polyfluoroarylation-Allylation of Diazo Compounds

Gao-Yin Wang et al.

Summary: Metal/ligand in situ assembly is crucial in transition metal catalysis for tailoring reactivity and selectivity. Cooperative catalysis via a single metal/two ligands is underdeveloped due to the challenge of harnessing distinct reactivity profiles of species generated by self-assembly. This study reports a catalytic system composed of a single metal/two ligands for a three-component reaction, leading to efficient construction of densely functionalized quaternary carbon centers.

Laura K. G. Ackerman-Biegasiewicz et al.

Summary: The use of two or more metal catalysts in a reaction is an efficient and selective synthetic strategy to access complex targets from simple starting materials. However, the principles governing multimetallic catalysis are not always intuitive, making the discovery and optimization of new reactions challenging. This article outlines the design elements of multimetallic catalysis using precedent from well-documented C-C bond-forming reactions, discussing advantages, limitations, and promoting the further development of the field.

Selective Construction of All-Carbon Quaternary Centers via Relay Catalysis of Indole C-H Functionalization/Allylic Alkylation

Jinyu Zhang et al.

Summary: A novel [Rh]2-catalyzed three-component reaction has been developed for the synthesis of C3 substituted indoles bearing all-carbon quaternary centers. This reaction involves a relay carbene-induced C-H functionalization and allylic alkylation process, showing good atom and step economy. Enantioselective control is achieved using a specific ligand, providing an efficient method for constructing enantioenriched indole derivatives with an all-carbon quaternary stereocenter.

Palladium-Catalyzed Three-Component Selective Aminoallylation of Diazo Compounds

Pengcheng Ou et al.

Summary: In this study, a Xantphos-containing dinuclear palladium complex was used to catalyze the geminal aminoallylation of diazocarbonyl compounds, providing a selective synthesis of various quaternary alpha-amino esters. The reaction did not proceed through direct N-H insertion, allylic alkylation of amino nucleophiles, or diene formation. Mechanistic investigations indicated that a relayed pathway via allylation or a [2,3]-sigmatropic rearrangement was unlikely.

Efficient Synthesis of Diaryl Quaternary Centers by Rh(II)/Xantphos Catalyzed Relay C-H Functionalization and Allylic Alkylation

Zhaoliang Ge et al.

Summary: A three-component reaction of N, N-disubstituted aniline, alpha-diazo ester, and an allylic electrophile has been achieved through [Rh(II)](2)/Xantphos catalysis, providing a direct access to diverse aniline derivatives with diaryl allylic quaternary centers in good yields. The synthetic utility of this method has been demonstrated by facile derivatization of the products, enabling the preparation of biologically relevant molecules and structural scaffolds, thereby offering a high potential for increasing molecular diversity. Mechanistic studies revealed the presence of a C(sp(2))-H functionalization of aniline derivatives/allylic alkylation cascade, with alpha, alpha-diarylacetate species identified as an active intermediate in this attractive catalytic transformation.
