Human cells with osteogenic potential in bone tissue research

Jana Dvorakova et al.

Summary: Bone regeneration is a challenging process in medicine, and various materials are being tested for replacing missing bone or teeth. Different cell types have their own advantages and limitations in the proliferation and differentiation of bone tissue. This review provides an overview of cell models, such as osteoblasts, mesenchymal stem cells, and osteosarcoma cells, for evaluating the biocompatibility of materials used in bone tissue research.

PF127 Hydrogel-Based Delivery of Exosomal CTNNB1 from Mesenchymal Stem Cells Induces Osteogenic Differentiation during the Repair of Alveolar Bone Defects

Longlong He et al.

Summary: This study demonstrated that CTNNB1-containing PF127 hydrogel effectively delivered bone marrow mesenchymal stem cell-derived exosomes, promoting osteogenic differentiation of the mesenchymal stem cells. Mechanistically, CTNNB1 regulated the miR-146a-5p/IRAK1/TRAF6 axis to induce the osteogenic differentiation process, leading to alveolar bone regeneration in rat models.

Epigenetic control of mesenchymal stem cells orchestrates bone regeneration

Xiaofeng Wang et al.

Summary: Recent studies have shown that MSCs play a vital role in bone regeneration, including osteogenic differentiation and providing seed cells. However, factors like drug intake and body senescence can affect MSCs' functions in bone regeneration. Epigenetic regulation has received attention for its heritable nature and involvement in bone metabolism disorders. This review focuses on the effects of epigenetic regulation on osteogenic differentiation, proliferation, and cellular senescence in MSCs and its role in bone regeneration.

The Use of Hydrogels for the Treatment of Bone Osteosarcoma via Localized Drug-Delivery and Tissue Regeneration: A Narrative Review

Shebin Tharakan et al.

Summary: Osteosarcoma, a malignant bone tumor, poses significant risks to patient mortality and morbidity. Conventional invasive treatments have drawbacks, prompting researchers to explore the use of hydrogels for targeted therapy. Hydrogels loaded with chemotherapeutic drugs have demonstrated promising results in eradicating tumor cells and promoting bone regeneration. Stimuli-responsive hydrogels can react to the tissue microenvironment, facilitating controlled drug release with modulated biomechanical properties. This narrative review discusses in vitro and in vivo studies on different hydrogels, including stimuli-responsive ones, for the treatment of osteosarcoma, as well as potential future applications for patient treatment.

Review on Strategies and Technologies for Exosome Isolation and Purification

Jiaci Chen et al.

Summary: Exosomes, as nano-sized extracellular vesicles, play an important role in cell-specific material transfer and have potential applications in prognosis, diagnosis, and treatment. Existing exosomal separation techniques have limitations in terms of complexity, time consumption, and low purity, highlighting the need for efficient and simple isolation methods. Microfluidic chips have emerged as a promising technology for exosome isolation. A comprehensive review covering exosomal properties, conventional technologies, emerging isolation strategies, and large-scale production of engineered exosomes is essential.

Clarissa Lenero et al.

Summary: This study discovered that endometrial mesenchymal stem/stromal cells (eMSC) and their CD146+ subpopulation possess exosomal secretomes with potent immunomodulatory miRNA attributes. These exosomes are involved in macrophage polarization, T cell activation, and regulation of inflammatory cytokines. The findings suggest that eMSC EXOs could be used for therapeutic purposes in resolving detrimental aspects of tissue inflammation.
