Immunogenicity of decellularized extracellular matrix scaffolds: a bottleneck in tissue engineering and regenerative medicine

Mohammadreza Kasravi et al.

Summary: Tissue-engineered decellularized extracellular matrix (ECM) scaffolds show promise in addressing donor shortage and immunologic rejection in tissue/organ transplantation. Decellularization removes immunogen cell materials and improves immunogenicity and biocompatibility of scaffolds. However, there are still major immunologic challenges in using these bioscaffolds.

Decellularized extracellular matrix scaffolds: Recent trends and emerging strategies in tissue engineering

Xuewei Zhang et al.

Summary: The application of decellularized extracellular matrix (dECM) scaffolds holds enormous potential for tissue regeneration. These scaffolds provide an optimal non-immune environment with native three-dimensional structures and bioactive components, making them an ideal choice for regenerating functional organs/tissues, especially when combined with stem cells.

Decellularized biologic muscle-fascia abdominal wall scaffold graft

Joseph F. Buell et al.

Summary: A novel decellularization method was successfully applied to create a DNA-free composite abdominal wall scaffold for intra-species implantation in rats without rejection. The scaffold combines the angiogenic capacity of decellularized muscle with the inherent strength of decellularized fascia, showing promising results for future preclinical analyses of graft strength in animal models.

Decellularization and In Vivo Recellularization of Abdominal Porcine Fascial Tissue

Julio C. Sanchez et al.

Summary: The decellularization protocol used in this study was efficient in removing cells and preserving the extracellular matrix of the fascial tissue. The mechanical testing showed that the decellularized samples maintained their physical properties. The successful transplantation of decellularized fascia into a rabbit and the absence of complications during a 6-month follow-up suggest that this biological scaffold could be a viable option for tissue engineering applications.

Biosynthesis of α-Gal Epitopes (Galα1-3Galβ1-4GlcNAc-R) and Their Unique Potential in Future α-Gal Therapies

Uri Galili

Summary: The alpha-gal epitope is a carbohydrate antigen synthesized by the glycosylation enzyme alpha 1,3galactosyltransferase in non-primate mammals, lemurs, and New-World monkeys, but extinct in ancestral Old-World monkeys and apes. The interaction between the alpha-gal epitope and anti-Gal in mammals can lead to hyperacute rejection during organ transplantation from non-primate mammals to Old-World monkeys or humans. Harnessing this interaction for development of novel immunotherapies (alpha-gal therapies) in various clinical settings may induce beneficial immune processes.

Abdominal wall reconstruction using biosynthetic absorbable mesh in high-risk complex ventral hernia

David Finch et al.

Summary: This study evaluated the performance of PGA:TMC biosynthetic mesh for reinforcement of midline fascial closure in high-risk patients undergoing complex ventral hernia repair. The results showed promising early outcomes, including significant improvements in quality of life and low hernia recurrence rate, associated with the use of biosynthetic mesh. Larger, well-controlled studies with longer follow-up are needed to confirm these findings.
