Reaction Behavior of [1,3-Diethyl-4,5-diphenyl-1H-imidazol-2-ylidene] Containing Gold(I/III) Complexes against Ingredients of the Cell Culture Medium and the Meaning on the Potential Use for Cancer Eradication Therapy

Paul Kapitza et al.

Summary: The reactivities of gold complexes with different ligands were analyzed in cell culture medium. Complexes 6 and 7 showed significant reactions, while interactions with non-thiol containing amino acids were not detected. Complex 8 exhibited strong stability and contributed to the biological effects of complex 7. The gold(III) species were reduced by GSH and showed promising activity against drug-resistant tumors.

Dibromo-Isonitrile and N-acyclic Carbene Complexes of Platinum(II): Synthesis and Reactivity

Anna Farasat et al.

Summary: A series of dibromo-N-acyclic (NAC) carbene complexes of platinum(II) were synthesized using a previously optimized protocol for analogous chlorinated compounds, starting from trans-[Pt(mu-Br)Br(PPh3)](2). The synthesis involved ring opening of the dinuclear precursor using suitable isonitrile ligands, followed by addition of N,N-diethylamine to the resulting products. Separate investigations were conducted for the two reactions with a focus on the differences between brominated and chlorinated systems.

trans-Dichloro(triphenylarsino)(N,N-dialkylamino)platinum(II) Complexes: In Search of New Scaffolds to Circumvent Cisplatin Resistance

Mariafrancesca Hyeraci et al.

Summary: Resistance to cisplatin limits its usefulness as an anticancer drug. This study describes the synthesis and characterization of two new platinum complexes and demonstrates their potential in overcoming cisplatin resistance in cancer cells. The complexes exhibited poor ability to platinate DNA but showed significant intracellular accumulation. Mechanistic studies revealed their ability to inhibit DNA relaxation and activate the apoptotic pathway. Comparison with related complexes provides insights into the resistance phenotype.

An overview of anticancer platinum N-heterocyclic carbene complexes

Summary: NHCs have been introduced as a new platform for the development of anticancer metal complexes, with platinum NHC complexes showing comparable activities to cisplatin and exhibiting different mechanisms of action, including the induction of biochemical hallmarks of immunogenic cell death (ICD) as a type II ICD inducer. These features make them promising candidates in the field of anticancer research.

New Platinum(II) Complexes Affecting Different Biomolecular Targets in Resistant Ovarian Carcinoma Cells

Mariafrancesca Hyeraci et al.

Summary: The new platinum complexes show significant antiproliferative effects on resistant ovarian carcinoma cells, demonstrating notable intracellular accumulation and the ability to interact with different intracellular targets. The mechanism of action involves interaction with DNA, collapse of mitochondrial transmembrane potential, and impairment of intracellular redox state, as well as selectivity towards selenocysteine of thioredoxin reductase.

Cytotoxicity and DNA interaction in a series of aryl terminated iminopyridine Pt(II) complexes

Riccardo Bondi et al.

Summary: The newly synthesized platinum(II) iminopyridine complexes exhibit good solubility and stability, with good interactions with DNA and significant antiproliferative activity on ovarian carcinoma cells. Complex 13 shows the ability to overcome cisplatin resistance and forms a dual geometry molecular complex with DNA, with hindered interstrand cross-links compared to cisplatin. The interactions with DNA are discussed in relation to the observed cell effects.
