Immobilisation of Cellobiose Dehydrogenase and Laccase on Chitosan Particles as a Multi-Enzymatic System for the Synthesis of Lactobionic Acid

Chitosan as a Promising Support of a CDH Activity Preservation System for Biomedical and Industrial Applications

Summary: The study aimed to immobilize cellobiose dehydrogenase (CDH) on chitosan beads and characterize the properties of immobilized CDHs from different fungal sources. The immobilized CDHs were characterized using FTIR spectra and SEM microstructure. The most effective method of immobilization was the covalent bonding of enzyme molecules using glutaraldehyde, showing efficiencies ranging from 28% to 99%. Promising results were obtained in terms of antioxidant, antimicrobial, and cytotoxic properties compared to free CDH. Chitosan appears to be a valuable material for the development of immobilization systems for biomedical applications or food packaging, while preserving the unique properties of CDH.

Characterization and Optimization of Injectable In Situ Crosslinked Chitosan-Genipin Hydrogels

Summary: Recently, there has been a growing interest in injectable, in situ crosslinking hydrogels that can conform to their environment with minimal invasion. Current in situ crosslinking chitosan hydrogels either lack biocompatibility and degrade slowly due to toxic crosslinking agents, or are mechanically weak and degrade rapidly due to insufficient crosslinking. In this study, the authors developed a thermally-driven, injectable chitosan-genipin hydrogel that can be crosslinked at 37 degrees C, which demonstrates mechanical strength, biodegradability, and high biocompatibility. The natural crosslinker used is genipin, which is non-toxic and activated by temperature. The characteristics of the chitosan-genipin hydrogel, such as crosslinking kinetics, injectability, viscoelasticity, swelling and pH response, and biocompatibility with keratinocyte cells, were analyzed. The results showed that the hydrogel can be successfully crosslinked at 37 degrees C, and it remains mechanically stable while being biodegradable. Long-term cell viability tests demonstrated excellent biocompatibility of the chitosan-genipin hydrogel, even during the crosslinking phase. Overall, these findings support the development of an injectable, in situ crosslinking chitosan-genipin hydrogel for minimally invasive biomedical applications.

Multi-Enzymatic Synthesis of Lactobionic Acid Using Wood-Degrading Enzymes Produced by White Rot Fungi

Summary: This study characterized a novel enzyme system for the synthesis of lactobionic acid (LBA) using enzymes from white rot fungi. The optimal conditions for LBA synthesis were found to be a temperature of 50 degrees C with the addition of ABTS as a redox mediator. The synthesized LBA showed excellent antioxidant and antibacterial properties, making it a promising compound for biotechnological applications.
