GelMA-glycol chitosan hydrogels for cartilage regeneration: The role of uniaxial mechanical stimulation in enhancing mechanical, adhesive, and biochemical properties

A Dual-Crosslinked Hydrogel Based on Gelatin Methacryloyl and Sulfhydrylated Chitosan for Promoting Wound Healing

Summary: This study designed a double-crosslinked hydrogel for wound dressing using photocrosslinkable gelatin methacryloyl (GelMA) and sulfhydrylated chitosan (CS-SH). The resulting hydrogels showed a highly porous inner structure, enhanced mechanical properties, and moisture retention capacity. In vitro studies showed no cytotoxicity and promoted cell proliferation. The double-crosslinked hydrogel also exhibited antibacterial properties and accelerated full-thickness skin defect healing in animal experiments.

Summary: Current surgical techniques for treating articular cartilage defects have limited long-term repair outcomes, while regenerative methods using biomaterials and stem cells show potential for generating hyaline cartilage. However, integrating the newly generated cartilage with surrounding tissue remains challenging. A strategy using a biological enzyme to bioadhere a gelatin methacryloyl implant to host tissue has been developed, offering improved mechanical properties and biocompatibility. This bioadhesive material can encapsulate cells, be photo crosslinked, and bond to host cartilage, representing a step towards integrating regenerative implants.
