Novel approaches to increase synaptic resilience as potential treatments for Alzheimer's disease

Summary: A significant proportion of cognitively normal patients with Alzheimer's disease pathology have differences in their brains that make them resilient to the disease. Recent approaches have focused on increasing synaptic resilience by targeting neurotrophic factors, protective genes, and proteins involved in dendritic spine strength. These strategies have the potential to treat Alzheimer's disease by making synapses resilient to its associated pathology, rather than targeting plaques and tangles.

In addition, combination therapies may be necessary for effective treatment of complex degenerative disorders.

Molecular Pathophysiological Mechanisms in Huntington's Disease

Summary: Huntington's disease is an inherited neurodegenerative disease that primarily affects medium spiny neurons in the striatum. The mutant huntingtin protein leads to neuronal loss through a series of molecular cascades, involving excitotoxicity, transcriptional dysregulation, and other key factors. In addition to neurons, astrocytes are also dysfunctional due to the presence of mutant huntingtin, and neuroinflammation may further contribute to the pathogenesis of Huntington's disease.
