Two weeks of high-intensity interval training increases skeletal muscle mitochondrial respiration via complex-specific remodeling in sedentary humans

Philip M. Batterson et al.

Summary: Aerobic training promotes substrate oxidation by remodeling the quantity and quality of skeletal muscle mitochondria. Short-term high-intensity interval training (HIIT) reveals changes to mitochondrial respiration and regulatory pathways. HIIT increases mitochondrial respiration for various substrates and induces gene transcripts and respiratory function, indicating remodeling of mitochondria within 2 weeks.

Exercise activates autophagy and regulates endoplasmic reticulum stress in muscle of high-fat diet mice to alleviate insulin resistance

Fangzhou Cheng et al.

Summary: This study investigates how exercise training acts on insulin resistance (IR) through the mechanism of endoplasmic reticulum (ER) stress and blocked autophagy caused by a high-fat diet (HFD). The findings suggest that exercise training can facilitate HFD-blocked protective autophagy via the activation of the AMPK/PGC1 alpha pathway to relieve insulin resistance in mice.

Sex-specific changes in metabolism during the transition from chow to high-fat diet feeding are abolished in response to dieting in C57BL/6J mice

Jennifer Oraha et al.

Summary: This study found that C57BL/6J mice exhibit sex-specific behavioral and metabolic responses to the introduction and sustained intake of a high-fat diet (HFD), but consistent responses to a dieting situation. Males showed immediate and significant weight gain, while females displayed stronger resistance to the obesifying effects of HFD. This sex disparity may be attributed to inherent sex differences in neurological signaling pathways and fat utilization. Interestingly, both sexes showed similar responses to returning to a normal diet.

Skeletal Muscle Nuclei in Mice are not Post-mitotic

Agnieszka K. Borowik et al.

Summary: The traditional belief that nuclei in skeletal muscle fibers are unable to replicate has been challenged by this study, which showed DNA synthesis and replication in resident myonuclei in vivo. The findings suggest that myonuclei have the potential to replicate and open up new avenues for muscle maintenance and growth.

Sex differences in changes of protein synthesis with rapamycin treatment are minimized when metformin is added to rapamycin

Christopher A. Wolff et al.

Summary: The study showed that Rapamycin and Rapamycin + Metformin have different effects on protein synthesis in young mice, with overall synthesis decreasing but individual synthesis rates varying. Metformin helps to balance the effects of Rapamycin on protein synthesis across different tissues and sexes, providing potential insights into how the combination may extend lifespan in both males and females.

Phenotypic sexual dimorphism in response to dietary fat manipulation in C57BL/6J mice

Summary: The study revealed significant differences in responses to high-fat diet between male and female mice, particularly in body weight gain, food intake, energy expenditure, and insulin sensitivity. Male mice gained weight faster, had an increase in pancreatic beta cell area, and reduced insulin sensitivity after HFD feeding, while female mice did not exhibit these phenomena.

Summary: The study found that a high-fat diet stimulates upstream activation of skeletal muscle mitophagy without altering mitochondrial respiration. This suggests that lipid-induced oxidative stress may be involved in this process.

Tumor burden negatively impacts protein turnover as a proteostatic process in noncancerous liver, heart, and muscle, but not brain

Jacob L. Brown et al.

Summary: Cancer survivors are more susceptible to pathologies such as hypertension, liver disease, depression, and coronary artery disease compared to those who have never been diagnosed with cancer. The study found that tumor burden can impact protein synthesis and proliferation rates in different tissues, with tumor tissue prioritizing protein synthesis for proliferation over proteostatic maintenance. Energetic stress was suggested as a contributing factor to the differences in tissue responses, highlighting the importance of proteostatic maintenance in liver, heart, and muscle in reducing disease risk for cancer survivors.

Determining the contributions of protein synthesis and breakdown to muscle atrophy requires non-steady-state equations

Kamil A. Kobak et al.

Summary: This study demonstrated the importance of accounting for a change in protein mass during D2O labelling studies, while also exploring the contribution of protein synthesis and breakdown to denervation-induced muscle atrophy. Conflicting results were found between steady- and non-steady-state calculations on myofibrillar protein synthesis and breakdown during periods of muscle loss. Additionally, collagen accumulation was largely from a decrease in collagen breakdown during denervation.

A Novel Stable Isotope Approach Demonstrates Surprising Degree of Age-Related Decline in Skeletal Muscle Collagen Proteostasis

Claire B. Abbott et al.

Summary: As age increases, turnover rate of collagen proteins slows down, accelerating extracellular matrix fibrosis and hindering adaptation to external stimuli. This research focused on understanding the factors that contribute to increased muscle fibrosis with age by studying the dynamic protein pool. Age-related decrease in the dynamic protein pool size in muscle collagen leads to increases in markers of fibrosis and decreased proteostatic maintenance.
