Role of the extracellular ATP/pyrophosphate metabolism cycle in vascular calcification

Ricardo Villa-Bellosta

Summary: ATP is traditionally considered as the main source of energy in cells, but recent studies have revealed its novel roles as a potent extracellular signaling molecule and the principal source of extracellular pyrophosphate, which is the main inhibitor of vascular calcification. High phosphate concentration and pyrophosphate deficiency are the principal mechanisms involved in ectopic calcification. Pathologic calcification of cardiovascular structures, known as vascular calcification, is a common complication of genetic diseases, chronic kidney disease, diabetes, and aging. Loss of function of enzymes and transporters involved in extracellular ATP/pyrophosphate metabolism has been associated with vascular calcification.

Radiologic evaluation and clinical effect of calcification in medial epicondylitis

Kyu Bok Kang et al.

Summary: This study revealed the characteristics and clinical relevance of calcification in medial epicondylitis (ME). The study found that calcification was more common in radiologic findings and had a relatively broad distribution. Calcification was associated with higher pain scores, a history of steroid injection, and combined ulnar neuropathy.

Rutin ameliorates the promotion effect of fine particulate matter on vascular calcification in calcifying vascular cells and ApoE-/- mice

Ruijun Su et al.

Summary: Atmospheric exposure to PM2.5 significantly contributes to the incidence and mortality of cardiovascular disease. This study reveals that PM2.5 promotes vascular calcification by enhancing hydroxyapatite deposition and activating the OPG/RANKL pathway. ROS generation induced by PM2.5 plays a crucial role in this process, and rutin effectively prevents vascular calcification by regulating the OPG/RANKL pathway.

Summary: This study compares two in vitro models of vascular smooth muscle cell (SMC) calcification, revealing distinct intracellular mechanisms triggered by OM and CaP. PKC plays a differential role in OM and CaP-induced SMC calcification, providing potential targets for pharmacological intervention in vascular calcification.

Vascular Calcification: Key Roles of Phosphate and Pyrophosphate

Ricardo Villa-Bellosta

Summary: Cardiovascular complications due to accelerated arterial stiffening and atherosclerosis are the leading cause of morbimortality in Western society, often associated with vascular calcification. The balance between phosphate and pyrophosphate, strictly controlled by several genes, plays a key role in calcification.

Evogliptin Suppresses Calcific Aortic Valve Disease by Attenuating Inflammation, Fibrosis, and Calcification

Bongkun Choi et al.

Summary: Evogliptin shows therapeutic potential in attenuating valvular calcification in CAVD animal models, reducing calcific deposition and inflammatory response in both in vivo and in vitro experiments. Additionally, it improves cardiac function and reduces valve calcification as assessed by echocardiography. In high-cholesterol diet and vitamin D2-driven fibrosis models, evogliptin suppresses inflammatory cell infiltration and calcification development in aortic valves.

Summary: TNF-alpha has specific effects on VIC phenotypes, potentially mediating calcification through the MAPK/ERK signaling cascade. VIC fibroblasts respond differently to TNF-alpha compared to profibrotic activated myofibroblasts, shedding light on the pathogenesis of valve disorders.

Summary: The study reveals that 5-MTP protects against atherosclerosis and calcification by inhibiting TLR2-mediated vascular smooth muscle cell phenotypic switch to chondrocytes and subsequent calcification. 5-MTP achieves these effects by blocking p38 MAPK activation and inhibiting CREB and NF-kappa B transactivation activity.
