Modifications of noncoding RNAs in cancer and their therapeutic implications

Summary: In the past 50 years, more than 150 chemical modifications on RNA molecules have been identified and characterized, playing important roles in regulating RNA biogenesis and biological functions. Among them, the epigenetic modification of noncoding RNAs (ncRNAs) has gained significant interest in the context of cancer due to the crucial roles of ncRNAs in cancer initiation and progression. This review summarizes various modifications of ncRNAs and highlights their potential as biomarkers and therapeutic targets in cancer.

JASPAR 2022: the 9th release of the open-access database of transcription factor binding profiles

CECR2 drives breast cancer metastasis by promoting NF-κB signaling and macrophage-mediated immune suppression

Summary: Transcriptomic analysis revealed that upregulation of CECR2 is associated with breast cancer metastasis and immune suppression. In mouse models, increased expression of CECR2 promotes breast cancer metastasis. Mechanistically, RELA, a member of the NF-kappa B family, recruits CECR2 to activate the expression of metastasis-related genes.

Functions of long non-coding RNA ROR in patient-derived glioblastoma cells

Summary: linc-RoR, a long non-coding RNA, plays an important role in glioblastoma. It is highly expressed in cells with increased EGFR expression. Knockdown of linc-RoR inhibits cell proliferation, increases sensitivity to DNA damage, and downregulates cancer stem cell markers. On the other hand, overexpression of linc-RoR promotes cell growth and increases the proportion of cancer stem cells. RNA sequencing analysis reveals that linc-RoR affects the expression of genes involved in mitotic regulation.

RELA is required for CD271 expression and stem-like characteristics in hypopharyngeal cancer

Summary: This study identified a novel long non-coding RNA (lncRNA) TMEM44-AS1 that promotes glioma progression by activating the Myc and EGR1/IL-6 signaling pathways, leading to increased proliferation, migration, and invasion of glioma cells through a positive feedback loop. A novel small-molecule Myc inhibitor, Myci975, was shown to alleviate the growth-promoting effects of TMEM44-AS1 in glioma cells, suggesting a potential anti-glioma therapeutic agent.
