The role of SLC transporters for brain health and disease

Yen T. K. Nguyen et al.

Summary: The brain exchanges nutrients and small molecules with blood through the blood-brain barrier, and glucose plays a critical role in providing energy for the brain. The brain also takes up various substances through different transport systems, and unraveling these mechanisms is key in understanding the brain's nutritional requirements and potential therapeutic interventions for neurological disorders.

Short chain fatty acids: Microbial metabolites for gut-brain axis signalling

Kenneth J. O'Riordan et al.

Summary: The role of the intestinal microbiota in regulating gut-brain axis signaling has gained attention in recent years. Understanding the relationship between the gut microbiota, its metabolites, and the brain is crucial for developing new therapeutic approaches, including identifying psychobiotics. Current research focuses on the effects of short-chain fatty acids (SCFAs), such as butyrate, acetate, and propionate, on brain function, behavior, and physiology, and how these SCFAs enter the systemic circulation and can potentially impact the brain.

Production of Indole and Indole-Related Compounds by the Intestinal Microbiota and Consequences for the Host: The Good, the Bad, and the Ugly

Naouel Tennoune et al.

Summary: The metabolic activity of intestinal microbiota generates various bacterial metabolites, including indole and indole-related compounds produced from tryptophan by specific bacterial species. These compounds have communication effects in the intestinal microbial community and generally beneficial effects on the intestinal mucosa. However, indole is partly metabolized into indoxyl sulfate in the liver, which can aggravate chronic kidney disease and cause endothelial dysfunction. Excessive concentrations of indole also have a negative impact on emotional behavior, while indole-derived co-metabolite isatin can modulate brain function positively or negatively. In addition, bacterial metabolite oxindole has deleterious effects on the central nervous system. The effects of indoxyl sulfate on the central nervous system vary depending on the dose used, with limited information on physiological concentrations.

Varied Composition and Underlying Mechanisms of Gut Microbiome in Neuroinflammation

Rai Khalid Farooq et al.

Summary: The gut microbiome has a significant impact on brain development and cognition, and neuroinflammation plays a role in the pathogenesis of neurological disorders. Restoring the gut microbiome through probiotics and fecal microbiome transplantation is important for reducing neuroinflammation.

Dietary fiber-based regulation of bile salt hydrolase activity in the gut microbiota and its relevance to human disease

Arthur Kastl et al.

Summary: This study investigates the effect of dietary fiber on patients with short bowel syndrome (SBS) and finds that SBS patients have lower diversity in their gut microbiota, which is inversely correlated with whole food consumption. Whole food intake is associated with increased glycoside hydrolases (GH) and bile salt hydrolases (BSH) and reduced fecal conjugated bile acids. Dietary fiber may regulate bile acid metabolism through the activity of gut microbiota enzymes, leading to intolerance of whole food in SBS patients.

Characterization of diet-dependent temporal changes in circulating short-chain fatty acid concentrations: A randomized crossover dietary trial

Jerusa Brignardello et al.

Summary: This study investigated the temporal changes in SCFA concentrations in response to two contrasting diets using a novel GC-MS method. The results showed that the unhealthy diet was associated with lower concentrations of certain SCFAs and higher concentrations of lactic acid. The study highlighted the importance of standardizing the timing of SCFA measurements in dietary studies.

Sodium butyrate ameliorates the impairment of synaptic plasticity by inhibiting the neuroinflammation in 5XFAD mice

Yu Jiang et al.

Summary: Research has shown that in the early stages of Alzheimer's disease progression, the use of sodium butyrate can reduce the secretion of pro-inflammatory cytokines by inhibiting histone deacetylase (HDAC), thereby improving synaptic plasticity in 5XFAD mice.

The phenotypic convergence between microglia and peripheral macrophages during development and neuroinflammation paves the way for new therapeutic perspectives

Francesca Grassivaro et al.

Summary: Microglia, the tissue resident macrophages of the brain, play a key role in central nervous system development and homeostasis. While specific markers have been identified for their investigation, recent studies have shown that their phenotype is constantly shaped by the microenvironment, leading to unexpected phenotypic convergence between microglia and peripheral macrophages at certain developmental stages and under pathological conditions. This novel information sheds new light on the therapeutic potential for neuroinflammatory and neurodegenerative diseases.
