Novel inhibitors of HSV-1 protease effective in vitro and in vivo

Magdalena Pachota et al.

Summary: Herpes simplex virus type 1 (HSV-1) is a common human pathogen causing infections of varying severity. This study discovered novel compounds that inhibit the activity of HSV-1 VP24 protease, effectively suppressing the infection in vitro and in vivo, including ACV-resistant strains. These inhibitors also prevent the nucleus egress of viral capsids and cell-to-cell spread. Due to their low toxicity and high antiviral potency, these VP24 inhibitors may serve as alternative therapeutics for ACV-resistant infections or be used in combination therapies.

Brilacidin, a COVID-19 drug candidate, demonstrates broad-spectrum antiviral activity against human coronaviruses OC43, 229E, and NL63 through targeting both the virus and the host cell

Yanmei Hu et al.

Summary: This study discovered the antiviral mechanism of Brilacidin, which inhibits the entry of multiple human coronaviruses by targeting HSPGs on the host cell surface. The study also found a strong synergistic effect of Brilacidin in combination with remdesivir against a specific coronavirus.

Self-Organized Nanoparticles of Random and Block Copolymers of Sodium 2-(Acrylamido)-2-methyl-1-propanesulfonate and Sodium 11-(Acrylamido)undecanoate as Safe and Effective Zika Virus Inhibitors

Pawel Botwina et al.

Summary: A series of polymers were synthesized and evaluated for their antiviral activity against Zika virus. The results showed that copolymers were more efficient antivirals compared to homopolymers, possibly due to their self-organization into nanoparticles that prevented virus attachment to host cells. Therefore, these copolymers could be promising agents against Zika virus infections.

Estimating excess mortality due to the COVID-19 pandemic: a systematic analysis of COVID-19-related mortality, 2020-21

Haidong Wang et al.

Summary: This study estimated excess mortality from the COVID-19 pandemic in 191 countries and territories, as well as 252 subnational units in selected countries from Jan 1, 2020, to Dec 31, 2021. The findings showed that globally, there were 18.2 million excess deaths due to the COVID-19 pandemic during this period. The highest excess mortality rates were observed in countries such as India, the USA, Russia, Mexico, Brazil, Indonesia, and Pakistan.

Spike-heparan sulfate interactions in SARS-CoV-2 infection

Fiona L. Kearns et al.

Summary: Heparan sulfate, a major component of the cellular glycocalyx, plays a crucial role in the infection cycle of SARS-CoV-2. It facilitates the open conformation of the spike protein, enabling its binding to ACE2. The coordination between viral and host glycans on the spike protein is essential for the binding and subsequent infection.

The glycocalyx affects the mechanotransductive perception of the topographical microenvironment

Matteo Chighizola et al.

Summary: This study used nano-bio-interfaces to analyze the impact of glycocalyx on PC12 cell mechanosensing at the nanoscale. The results showed that the glycocalyx configuration affects mechanotransductive events at the cell/microenvironment interface, with glycocalyx removal leading to reduced retrograde actin flow speed and force loading on certain nanotopographies.

The in vitro antiviral activity of lactoferrin against common human coronaviruses and SARS-CoV-2 is mediated by targeting the heparan sulfate co-receptor

Yanmei Hu et al.

Summary: The study highlights the important role of HSPGs in SARS-CoV-2 cell attachment, and demonstrates the broad-spectrum antiviral activity of LF against various coronaviruses, especially with BLF being more potent than HLF. BLF binds to HSPGs to block viral attachment, and its antiviral activity can be antagonized by the HSPG mimetic heparin. Combination therapy experiment shows synergistic effect of LF with remdesivir in cell culture.
