Remodeling liver microenvironment by L-arginine loaded hollow polydopamine nanoparticles for liver cirrhosis treatment

Yeying Wang et al.

Summary: Liver cirrhosis is a liver disease with a high mortality rate worldwide. Antifibrotic drugs are commonly used clinically, but there are still many challenges. Excessive reactive oxygen species (ROS) in the liver lesions microenvironment is an important factor leading to the development of liver cirrhosis. Therefore, a nanomedicine-mediated antioxidant therapy can remodel the liver microenvironment and reverse the process of cirrhosis.

Camouflaged Virus-Like-Nanocarrier with a Transformable Rough Surface for Boosting Drug Delivery

Hongyue Yu et al.

Summary: A camouflaged virus-like nanocarrier with a transformable rough surface composed of a mesoporous SiO2 nanoparticle and an acid-responsive polymer is reported. Under normal pH conditions, the spikes on the nanocarrier are hidden by the polymer shell, inhibiting non-specific strong nano-bio interactions. In an acidic tumor microenvironment, the nanocarrier sheds the polymer camouflage to expose its rough surface, improving retention ability and endocytosis efficiency. The rationally designed nanocarrier exhibits extended blood circulation time and enhanced tumor accumulation due to its dynamically variable rough surface.

Immunogenic cell stress and death

Guido Kroemer et al.

Summary: Immunogenic cell death (ICD) plays an important role in therapy and disease, as dying mammalian cells release signals that interact with the host to determine the immunological correlates of cellular stress and death. ICD is crucial in immunosurveillance and is related to strategies evolved by pathogens and cancer cells. Additionally, normal cell death can also initiate antigen-specific immune responses.

Kesi Wang et al.

Summary: In this study, a nano-vesicular system IA-PEP was developed to efficiently deliver photosensitizer PEGylated indocyanine green and NO donor L-Arg, achieving a ternary synergistic treatment combining NO therapy, photodynamic therapy, and mild-temperature photothermal therapy under 808 nm laser irradiation. The potent anti-tumor efficacy of IA-PEP with complete tumor elimination was verified in vivo in nude mice bearing xenograft MCF-7 tumors.

A responsive microneedle system for efficient anti-melanoma by combining self-enhanced chemodynamic therapy with photothermal therapy

Jiajie Chen et al.

Summary: Traditional therapies for melanoma have limitations, which necessitates the development of new treatments. The use of a responsive microneedle system that combines self-enhanced chemodynamic therapy and photothermal therapy has shown promise in effectively treating melanoma. The system releases CuO2 nanoparticles that generate reactive oxygen species in response to the acidic tumor environment, while also inducing local heat for photothermal therapy.

Specific generation of nitric oxide in mitochondria of cancer cell for selective oncotherapy

Mengyu Sun et al.

Summary: A nanozyme-based NO generator, cerium oxide (CeO2)-AT, has been developed to selectively produce nitric oxide (NO) in cancer cells for tumor treatment. The generator can disrupt the mitochondrial respiratory chain of tumor cells and induce cell apoptosis by catalyzing the production of NO in the mitochondria of cancer cells. Furthermore, the generator can improve the performance of NO gas therapy by catalyzing H2O2 to produce O-2. This study provides a new approach for safe and specific cancer therapy using highly selective NO-based treatment.

Targeting tumor-associated macrophages for cancer treatment

Mengjun Li et al.

Summary: Tumor-associated macrophages (TAMs) play a crucial role in the tumor microenvironment due to their immunosuppressive and other functions, making them important targets for anti-cancer strategies. However, the heterogeneity of TAMs makes the targeting strategy variable and uncertain. Identifying the subset specificity of TAMs may be a future option for treatment.
