Strategies for Supplying Precursors to Enhance the Production of Secondary Metabolites in Solid-State Fermentation

Enhancing bioflavor production by solid-state fermentation using Kluyveromyces marxianus and l-phenylalanine

Summary: This study explores the production of bioflavor compounds, specifically 2-phenylethanol (2-PE) and 2-phenylethyl acetate (2-PEA), using Kluyveromyces marxianus in solid-state fermentation with sweet lemon peel (SLP) and sugarcane bagasse (SB). The quantification of flavor compounds revealed that without any supplements, the concentrations of 2-PE were 0.15 +/- 0.003 mg/g for SLP and 0.14 +/- 0.002 mg/g for SB, while the concentrations of 2-PEA were 0.01 +/- 0.008 mg/g for SLP and 0.02 +/- 0.001 mg/g for SB. The addition of l-phenylalanine (l-phe) showed a 30%-75% enhancement in the production of both 2-PE and 2-PEA. This study suggests that K. marxianus, with the addition of synthetic l-phe, is a promising microbial cell factory for the production of 2-PE and 2-PEA, which can be applied in various industries such as food and pharmaceutical.

However, natural foods usually have low levels of GABA, and using enrichment technologies to increase GABA content instead of exogenous addition can enhance consumer acceptance among health-conscious individuals.

Pilot-scale process to produce bio-pigment from Monascus purpureus using broken rice as substrate for solid-state fermentation

Summary: This study successfully produced natural red bio-pigment at a pilot-scale by optimizing fermenter type and bio-pigment extraction parameters. The tray-type fermenter outperformed other types for bio-pigment production. The high yield of bio-pigment was extracted with 60% ethanol at 60°C for 80 min. The study also found three color forms of bio-pigment in the extract, namely red, orange, and yellow. The extracted red bio-pigment exhibited sensitivity to UV, sunlight, and pH > 10, but could withstand high temperature and low concentrations of food preservatives.

Summary: Ochratoxin A is a toxic secondary metabolite produced by Aspergillus ochraceus. The identification and prediction of diverse compounds and their biosynthesis pose a challenge. Previous studies in Europe and the USA failed to produce ochratoxin A in certain bean isolates. Analysis of metabolites led to the discovery of notoamide R and its biosynthetic components in a shredded-wheat/shaken-flask fermentation. Further investigations confirmed notoamide R as a prominent metabolite of A. ochraceus. The study also highlighted the importance of intermediary metabolism in secondary metabolite accumulation.

CRISPR/Cas9-mediated genome editing in vancomycin-producing strain Amycolatopsis keratiniphila

Summary: Amycolatopsis is a valuable source of bioactive natural products, and this study aimed to develop a high-efficiency CRISPR/Cas9-mediated genome editing system in the vancomycin-producing strain A. keratiniphila HCCB10007. The researchers successfully utilized an all-in-one editing plasmid to delete and insert specific genes with 100% efficiency. They also achieved high efficiencies of 81%-97% in deleting large fragments of the ECO-0501 biosynthetic gene cluster by selecting suitable sgRNAs. The deletion of these gene clusters led to a significant improvement in vancomycin production.

Strategy for Producing the High-Quality Glycopeptide Antibiotic A82846B in Amycolatopsis orientalis Based on the CRISPR-Cas12a System
