Design and application of artificial rare L-lysine codons in Corynebacterium glutamicum

Summary: In this study, high L-lysine-producing strains were screened by constructing the artificial rare codon AAA in C. glutamicum. Fermentation results showed that the mutant strains with higher fluorescence intensities had an increase of up to 9.7% in L-lysine production, and the highest screening positive rate was 69%. Therefore, the application of artificially constructed rare codons provides an efficient, accurate, and simple method for screening other amino acid-producing microorganisms.

Biosensor-assisted evolution for high-level production of 4-hydroxyphe- nylacetic acid in Escherichia coli

Summary: In this study, a biosensor for 4-hydroxyphenylacetic acid (4HPAA) was used to identify preferred mutants for higher 4HPAA production and tolerance. The mutant strain GS-2-4, obtained through atmospheric and room temperature plasma mutagenesis coupled with adaptive laboratory evolution, produced 25.42 g/L 4HPAA. Through whole genome sequencing, chromosomal mutations of GS-2-4 were identified. Important genes, pathways, and regulations involved in 4HPAA biosynthesis were revealed. The combination of acrD-aroG further increased 4HPAA production of GS-2-4 by 37%.

Escherichia coli BW25113 Competent Cells Prepared Using a Simple Chemical Method Have Unmatched Transformation and Cloning Efficiencies

Summary: This study found that using E.coli BW25113 (recA(+)) competent cells can achieve higher transformation efficiency and cloning success rate compared to commonly used E.coli cloning strains, with RecA playing a partial role in the differences. Even with the recA deletion in BW25113, the transformation efficiency and cloning success rate are still significantly higher than those of commonly used cloning strains. These findings suggest that E.coli BW25113 is an ideal choice for efficient cloning and DNA assembly.
