Neuronal activity-induced, equilibrative nucleoside transporter-dependent, somatodendritic adenosine release revealed by a GRAB sensor

Zhaofa Wu et al.

Summary: Using a genetically encoded GPCR-Activation-Based Ado fluorescent sensor, researchers found that extracellular Ado elevation induced by neuronal activity in the hippocampus is due to direct Ado release from neuronal somatodendritic compartments, rather than from axonal terminals. The release of Ado depends on equilibrative nucleoside transporters and is slow and calcium-dependent.

Using mass spectrometry imaging to map fluxes quantitatively in the tumor ecosystem

Michaela Schwaiger-Haber et al.

Summary: Tumors are heterogeneous and consist of various cell types in different microenvironments. The integration of mass spectrometry imaging (MSI), stable isotope labeling, and isotopologue spectral analysis enables the mapping of metabolite distributions and metabolic fluxes in the brains of mice with GL261 glioma. This approach reveals alterations in anabolic pathways, specifically an increase in de novo fatty acid synthesis and elongation fluxes in the glioma compared to surrounding healthy tissue.

HMDB 5.0: the Human Metabolome Database for 2022

David S. Wishart et al.

Summary: The Human Metabolome Database (HMDB) has been providing comprehensive information about human metabolites since 2007, and has undergone significant improvements and upgrades in its latest update, HMDB 5.0. These improvements include an increase in the number of metabolite entries, enhancements to metabolite descriptions, new visualization tools, and more accurately predicted spectral data sets. These upgrades are aimed at improving the usability and potential applications of the HMDB in various fields, including human metabolomics, exposomics, lipidomics, nutritional science, biochemistry, and clinical chemistry.

Comparison of Cosine, Modified Cosine, and Neutral Loss Based Spectrum Alignment For Discovery of Structurally Related Molecules

Wout Bittremieux et al.

Summary: A comparative analysis was performed to evaluate the scoring properties of neutral loss matching, cosine similarity, and modified cosine similarity in MS/MS data. The results showed that the modified cosine similarity outperformed the other methods in all cases, and the performance of MS/MS spectrum alignment depended on the location, type, and chemical compound class of modifications.

Spatially resolved characterization of tissue metabolic compartments in fasted and high-fat diet livers

Sylwia A. Stopka et al.

Summary: Cells adapt their metabolism in response to physiological cues, and metabolic heterogeneity exists at various levels. The liver plays a crucial role in maintaining overall metabolic balance, and its structure is defined by metabolic zones. Mass spectrometry imaging enables the mapping of metabolites directly from tissue sections, allowing for a better understanding of metabolic heterogeneity compared to traditional methods. This study establishes a workflow for matrix-assisted laser desorption/ionization mass spectrometry imaging and provides insights into liver metabolism under nutrient stress and excess.

Purine nucleoside phosphorylase enables dual metabolic checkpoints that prevent T cell immunodeficiency and TLR7-associated autoimmunity

Evan R. Abt et al.

Summary: The deficiency of PNP can lead to both immunodeficiency and autoimmunity. The mechanisms behind this paradoxical association are not fully understood. This study reveals that the inactivation of PNP can affect T cell development and B cell signaling, resulting in immune abnormalities and autoimmune diseases.

Analyzing cell-type-specific dynamics of metabolism in kidney repair

Gangqi Wang et al.

Summary: In this study, the authors used mass spectrometry imaging, isotope tracing, and multiplexed immunofluorescence microscopy to study metabolic dynamics in the kidney during ischemia-reperfusion. They developed a high-spatial-resolution metabolomics approach that allows mapping of cell-type-specific changes in central carbon metabolism. This method, combined with immunofluorescence staining, can detect metabolic changes and nutrient partitioning in targeted cell types. The authors identified region-specific metabolic perturbations associated with the lesion and throughout recovery, providing insights into the homeostatic capacity of the kidney microenvironment.

Stop the rot. Enzyme inactivation at brain harvest prevents artifacts A guide for preservation of the in vivo concentrations of brain constituents

Gerald A. Dienel

Summary: Post-mortem metabolism causes rapid and prolonged changes in brain concentrations of various compounds, which are labile. Properly stopping metabolism at harvest and reporting absolute concentration units are crucial for improving metabolomic studies of the brain. These methods can enhance understanding of both normal and diseased brain conditions, while also preventing the reporting of artifactual data.

Raf promotes dimerization of the Ras G-domain with increased allosteric connections

Morgan R. Packer et al.

Summary: Studies demonstrate that Raf-Ras binding domain (Raf-RBD) can induce Ras dimerization at low surface densities and in solution, with robust connections between two Raf-RBD D113 residues confirmed by molecular dynamics simulations. These results suggest that Raf-RBD binding and Ras dimerization occur together, forming a high-affinity signaling complex.
