Biomaterial Strategies for Selective Immune Tolerance: Advances and Gaps

Lipid-Polymer Hybrid Nanoparticles Utilize B Cells and Dendritic Cells to Elicit Distinct Antigen-Specific CD4+ and CD8+ T Cell Responses

Summary: Antigen-presenting cells (APCs) play a crucial role in treating immune-mediated diseases, but their clinical translation is limited by challenges such as control of antigen dosing and low abundance. This study explores the use of B cells as an alternative to dendritic cells (DCs) for T cell priming, focusing on the development of delivery platforms using phospholipid-conjugated antigens (L-Ags) and lipid-polymer hybrid nanoparticles (L/P-Ag NPs). The results show that L-Ag delivery effectively loaded all types of APCs and primed antigen-specific CD8(+) and CD4(+) T cells. The incorporation of L-Ags and polymer-conjugated antigens into nanoparticles allowed for different antigen uptake pathways, shaping T cell responses in DCs and B cells. This research provides a modular delivery platform for engineering antigen-specific immunotherapies.

Mechanistic contributions of Kupffer cells and liver sinusoidal endothelial cells in nanoparticle-induced antigen-specific immune tolerance

This mechanistic support provides insights for the development of biomaterial platforms in clinical trials.
