Iron oxide@chlorophyll clustered nanoparticles eliminate bladder cancer by photodynamic immunotherapy-initiated ferroptosis and immunostimulation

Summary: Designing a multifunctional nanoparticle that boosts programmed cell death and immunoactivation has potential as a treatment strategy for bladder cancer. In this study, cluster-structured nanoparticles were developed that could perform fluorescence, change the redox balance, and attenuate lipid peroxidation. Targeting the bladder wall and inducing cytotoxicity and ferroptosis resulted in a reprogramming of the tumor microenvironment from immunosuppressive to immunostimulatory. Intravesical instillation of these nanoparticles improved antitumor efficacy and survival rates.

Summary: The facile and straightforward fabrication of NIR-responsive theranostic materials with high biocompatibility is still an unmet need for nanomedicine applications. Here, we used a natural photosensitizer, iron chlorophyll (Chl/Fe), for the J-aggregate template-assisted synthesis of Au@ Chl/Fe nanorods with high stability. The assembly of a high amount of Chl/Fe J-aggregate onto the Au surface enabled red-NIR fluorescence for monitoring and tracking residential tumor lesions.

Fibrillation of Human Calcitonin and Its Analogs: Effects of Phosphorylation and Disulfide Reduction

Summary: The study demonstrates the potential anti-amyloidogenic properties of endogenously derived compounds based on porphyrin, which can inhibit fibril formation of insulin and hen egg white lysozyme. Hematoporphyrin, derived from heme by hydroxylation and metal removal, shows superior inhibitory effects on insulin fibril formation compared to hemin and protoporphyrin.

Inhibition of tau amyloid formation and disruption of its preformed fibrils by Naphthoquinone-Dopamine hybrid

Summary: The dopamine-based hybrid molecule NQDA effectively inhibits the aggregation of key tau-derived amyloidogenic fragments PHF6 and PHF6*, as well as full-length tau protein, in in vitro experiments. It disassembles preformed fibrils and demonstrates significant binding efficiency with tau fibrils through entropic and enthalpic processes. NQDA shows comparable or lower IC50 values for inhibiting fibril formation compared to other amyloid inhibitors, suggesting its potential as a scaffold for developing therapeutics for Alzheimer's disease and other tauopathies.
