Summary: In 2022, icaritin soft capsules were approved by the NMPA of China for the treatment of HCC. However, the low content, poor bioavailability, and unfavorable delivery efficiency of epimedium flavonoids have hampered their production efficiency and clinical applications. Various strategies, including enzyme engineering and nanotechnology, have recently been developed to increase productivity, improve delivery efficiency, and enhance therapeutic effects. This review discusses the structure-activity relationship of epimedium flavonoids, enzymatic engineering strategies, and nanomedicines for overcoming delivery barriers and improving therapeutic effects.

Dual Nanozyme-Driven PtSn Bimetallic Nanoclusters for Metal-Enhanced Tumor Photothermal and Catalytic Therapy

Summary: Generating reactive oxygen species (ROS) within tumors using nanozymes is a promising strategy for cancer therapeutics. Here, a bimetallic nanozyme (Pt50Sn50) with photothermal enhancement of enzymatic activities for tumor catalytic therapy is developed. The Pt50Sn50 nanozyme exhibits the highest activities among all investigated due to the generation of SnO2-x with oxygen vacancy sites on its surface.

Ultra-Small and Metabolizable Near-Infrared Au/Gd Nanoclusters for Targeted FL/MRI Imaging and Cancer Theranostics

Summary: This study engineered ultra-small and metabolizable dual-mode imaging probe Au/Gd@FA NCs using a simple hydrothermal method, which showed superior properties compared to commercial MRI contrast agents. The probes exhibited high quantum yield, near-infrared fluorescence, and a better T-1 nuclear magnetic signal. By modifying the folic acid molecules, the probes achieved specific fluorescence imaging of breast cancer. Moreover, the probes were deemed safe and could promote intracellular ROS production and induce cell apoptosis. This metabolizable probe provides a potential candidate for multimodal imaging and tumor diagnosis in basic research.
