Organic Photothermal Cocrystals: Rational Design, Controlled Synthesis, and Advanced Application

Summary: Organic photothermal cocrystals, which combine the advantages of organic cocrystals and photothermal conversion ability, have attracted a lot of attention in various fields such as photoacoustic imaging, seawater desalination, and photothermal therapy. However, there is still a lack of systematic investigation into the photothermal conversion mechanisms of these cocrystals, posing a challenge for further developments. Therefore, it is crucial to explore the rational design and synthesis of high-performance organic photothermal cocrystals for future applications. This review provides a comprehensive summary of the molecular classification, photothermal conversion mechanisms, and applications of organic photothermal cocrystals.

Advances in Single-component inorganic nanostructures for photoacoustic imaging guided photothermal therapy

Summary: Phototheranostic based on photothermal therapy (PTT) and photoacoustic imaging (PAI) is gaining attention due to its noninvasive, deeply penetrative, and highly selective and effective therapy. Single-component inorganic nanostructures are novel and attractive PAI and PTT combined nanotheranostic agents. They have controllable structure, tunable physiochemical properties, size-dependent optical properties, simple composition, and easy synthesis process. They also have multiple therapeutic and imaging functions, reducing systemic side effects and improving the bioavailability.

Structure-Property-Function Relationships of Iron Oxide Multicore Nanoflowers in Magnetic Hyperthermia and Photothermia

Summary: Magnetite and maghemite multicore nanoflowers synthesized using modified polyol-mediated routes are highly effective nanoheaters in magnetic hyperthermia (MHT) and have also shown promising performances in photothermal therapy (PT) in the NIR-II biological window. The study investigates the influence of synthesis parameters on the size, shape, composition, cores, and crystallinity of the nanoflowers, establishing correlations between their structure, properties, and function in MHT and PT. Iron(III) and heating ramps are found to play a critical role in the formation of well-defined nanoflowers with multiple cores, with optical responses and photothermal properties depending on volume and structural disorder rather than stoichiometry. Defect concentration in the nanostructures affects optical behavior, with a transition in photothermal efficiency observed at a critical Urbach energy value.

Collective Plasmon Coupling in Gold Nanoparticle Clusters for Highly Efficient Photothermal Therapy

An Ultrastable 155-Nuclei Silver Nanocluster Protected by Thiacalix[4]arene and Cyclohexanethiol for Photothermal Conversion

The structure consists of a metallic core with four concentric shells, surrounded by an organic skin. This work not only reveals the influence of macrocyclic ligands on the formation of large silver nanoclusters, but also provides a new structural archetype.

Heat-Generating Mn0.5Zn0.5Fe2O4/MWCNTs nanocomposites for enhancing hyperthermia efficacy in magnetic hyperthermia applications

Summary: In this study, Mn0.5Zn0.5Fe2O4/MWCNTs nanocomposites were synthesized using the chemical co-precipitation method for cancer treatment. The addition of a small amount of MWCNTs resulted in increased saturation magnetization. The nanocomposites exhibited good magnetic hyperthermia properties and could achieve suitable temperatures for tumor treatment at lower frequencies, while also showing good biocompatibility.

pH/Thermal-Sensitive Nanoplatform Capable of On-Demand Specific Release to Potentiate Drug Delivery and Combinational Hyperthermia/Chemo/Chemodynamic Therapy

Summary: A versatile nanoplatform, PDDFe, capable of tumorspecific delivery and controlled drug release was constructed. The nanoplatform utilized iron-oxide nanoparticles and doxorubicin encapsulated in pH/thermal-sensitive micelles to achieve remarkable magnetic targeting effects and specific accumulation at tumor locations. The release of the drug was triggered by the acidic environment and localized heat generated by hyperthermia therapy, leading to a pronounced therapeutic effect. In vivo imaging experiments validated the site-specific accumulation of the nanoplatforms at the tumor sites. The combination of PDDFe with alternating magnetic field and laser irradiation effectively retarded tumor growth through hyperthermia/chemo/chemodynamic therapy.

Gold Nanocage-Based Photothermal Ablation Facilitates In Situ Vaccination for Melanoma Therapy

Summary: This study developed an in situ vaccination strategy using gold nanocage (AuNC)-based photothermal effect in combination with adjuvant and PD-L1 suppressor to alleviate intratumoral immunosuppression in cancer immunotherapy. The results showed that this strategy effectively activated immune cells and demonstrated high therapeutic efficacy in a melanoma mouse model. It increased infiltration of cytotoxic T lymphocytes, suppressed PD-L1 expression, and repolarized tumor-associated macrophages, thereby remodeling the tumor microenvironment.

Summary: Nanosystems containing both fluorescent and magnetic modules have great potential in biomedical applications. In this study, a magneto-luminescent system based on Fe3O4 nanoparticles and thermosensitive rhodamine fluorophores was designed, which can self-monitor the degree of hyperthermia. The optimized nanoparticles show suitable fluorescence thermosensitivity and excellent heating capacity within the clinical safety limits, opening up promising avenues for next-generation medical technologies.
