Selective Accumulation to Tumor Cells with Coacervate Droplets Formed from a Water-Insoluble Acrylate Polymer

Kei Nishida et al.

Summary: Coacervate droplets formed from poly(2-methoxyethyl acrylate) (PMEA) and its derivatives selectively accumulate to tumor cells via hydrophobic interaction. The accumulation of droplets is influenced by the bound water content of PMEA derivatives and the interactions with plasma membrane components, such as CD44. These coacervate droplets have great potential in tumor cell detection, development of alternative ligands, and drug delivery optimization.

Critical clinical gaps in cancer precision nanomedicine development

Wendy Wei Gan et al.

Summary: Active targeting strategy is used in nanomedicine for cancer treatment. Personalization of nanomedicine based on patients' omics profiles faces challenges in ligand and matrix material selection. Past research shows that the selection of nanoparticulate ligands and matrix materials does not consider patients' cancer omics. Omics analysis of individual metabolizing enzymes and cancer cell surface receptors can aid in the selection of ligands and matrix materials for nanomedicine development.

A targeted nanoplatform co-delivery of pooled siRNA and doxorubicin for reversing of multidrug resistance in breast cancer

Hongmei Liu et al.

Summary: This study aimed to develop a novel and facile strategy for preparing a highly efficient and safe gene and drug delivery system for multidrug-resistant (MDR) breast cancer. A targeted nanoplatform based on biodegradable boronic acid modified epsilon-polylysine was developed to co-deliver P-gp siRNA, Bcl-2 siRNA, and doxorubicin for overcoming MDR. The nanoplatform exhibited robust inhibitory effects against invasion, proliferation, and colony formation of MCF-7/ADR cells in vitro, and demonstrated ATP responsiveness. In vivo antitumor experiments showed significant inhibition of tumor growth and good biocompatibility with the targeted nanoplatform.

Liposomal Co-delivery of PD-L1 siRNA/Anemoside B4 for Enhanced Combinational Immunotherapeutic Effect

Xiang Li et al.

Summary: Combination therapy using cRGD-targeted liposomal preparation for co-delivery of PD-L1 siRNA and AB4 showed promising anticancer efficiency in mouse models. The liposomes exhibited good drug encapsulation, pH-sensitive release properties, and stable characteristics. The combination treatment downregulated PD-L1 protein expression, modulated the immunosuppressive microenvironment, and showed no obvious toxicity. This approach may provide a foundation for the development of a new targeted drug delivery system.
