Computational NMR of Carbohydrates: Theoretical Background, Applications, and Perspectives

Summary: This review highlights the significant progress in the calculation of NMR parameters of carbohydrates, supported by a vast amount of experimental data from various laboratories worldwide. The main purpose of this review is to outline general trends and perspectives in this dynamically developing area against the background of marked progress in theoretical and computational NMR. The presented material is divided into three basic sections, covering theoretical introduction, applications and perspectives in computational NMR of monosaccharides, as well as calculation of NMR chemical shifts and spin-spin coupling constants of di- and polysaccharides.

The use of MM/QM calculations of 13C chemical shifts in the conformational analysis of some monosaccharides and sucrose

Summary: A combined molecular mechanics and ab initio model was utilized to predict C-13 NMR chemical shifts of common pyranose monosaccharides in D2O solution, showing good agreement with experimental results. The populations of conformers at C-6 of the monosaccharides were obtained, revealing similar ratios between alpha and beta anomers in all compounds studied. This study represents the first utilization of C-13 shifts to determine sugar conformations in solution, providing valuable insights into the structural preferences of different monosaccharides.
