Biased Agonism: Lessons from Studies of Opioid Receptor Agonists

Summary: In ligand bias, different agonist drugs activate distinct signaling pathways, leading to different therapeutic and adverse effects. While it was believed that selectively activating the G protein-dependent pathway of the mu-opioid receptor would result in effective analgesia without adverse effects, recent data suggest that most effects are mediated by this pathway and some drugs described as biased may actually be low-intrinsic-efficacy agonists. This review discusses the current understanding of bias at the mu-opioid receptor and other opioid receptor subtypes.

Discovery of a Potent Highly Biased MOR Partial Agonist among Diastereomeric C9-Hydroxyalkyl-5-phenylmorphans

Summary: All possible diastereomeric C9-hydroxymethyl-, hydroxyethyl-, and hydroxypropyl-substituted 5-phenylmorphans were synthesized to explore the three-dimensional space around the C9 substituent in search for potent MOR partial agonists. Many of these compounds exhibited nanomolar or subnanomolar potency and were fully efficacious. One compound in particular, 21, showed good potency and efficacy as a MOR partial agonist, as well as selectively activating G-protein signaling pathway. Overall, the study highlights the importance of structural modification in developing new opioids with improved analgesic properties.

Summary: The degree of mu opioid receptor (MOR) efficacy required to produce hyperactivity in mice was evaluated in this study. The results showed that the hyperactivity induced by MOR agonists is efficacy dependent and requires relatively high levels of MOR agonist efficacy for its full expression.

Opioid-like adverse effects of tianeptine in male rats and mice

Summary: The study found that tianeptine has a lower abuse potential compared to commonly abused opioids in rats, as it exhibits resistance to tolerance and dependence. However, tianeptine does produce acute adverse effects similar to opioid agonists, including motor impairment, constipation, and respiratory depression.

The study found similarities and differences in their functional activities and pharmacological properties, and identified potential structures that contribute to these differences through molecular modeling.

Manipulating Pharmacodynamic Efficacy with Agonist plus Antagonist Mixtures: In Vitro and In Vivo Studies with Opioids and Cannabinoids

Summary: The pharmacodynamic efficacy of drugs in activating their receptors is crucial in determining drug effects. Fixed-proportion mixtures of an agonist and antagonist for a given receptor can be adjusted to precisely control net efficacy of the mixture in activating that receptor. Manipulation of agonist proportion in agonist/antagonist mixtures governs net mixture efficacy at the target receptor.
