Metabolic functional redundancy of the CYP9A subfamily members leads to P450-mediated lambda-cyhalothrin resistance in Cydia pomonella

Pei-Rong Li et al.

Summary: The study found that functional redundancy and preference of metabolism by CYP9A genes in the CYP9A subfamily contribute to lambda-cyhalothrin resistance in Cydia pomonella. These genes are highly expressed in the larval midgut and are induced by lambda-cyhalothrin. Knockdown experiments and in vitro assays confirmed the metabolic roles of these genes in lambda-cyhalothrin. These findings reveal the molecular mechanism of P450-mediated lambda-cyhalothrin resistance and its adaptive evolutionary strategy in Cydia pomonella.

Synergistic effects of chlorantraniliprole and camptothecin on physiological impairments, histopathological, biochemical changes, and genes responses in the larvae midgut of Spodoptera frugiperda

Patrick Maada Ngegba et al.

Summary: Spodoptera frugiperda is a major agricultural pest that threatens global food security. It is highly polyphagous, migratory, and fecund, making its management challenging. The insecticide chlorantraniliprole is commonly used to control S. frugiperda, but its overuse poses health risks, resistance development, toxicity to beneficial insects, and environmental contamination. This study investigates the synergistic effects of camptothecin with chlorantraniliprole in managing S. frugiperda resistance and associated challenges.

Glutathione S-Transferase Genes are Involved in Lambda-Cyhalothrin Resistance in Cydia pomonella via Sequestration

Chao Hu et al.

Summary: This study reports the involvement of GSTs in lambda-cyhalothrin resistance in the globally quarantined fruit pest Cydia pomonella. Most GST genes were upregulated in response to exposure to lambda-cyhalothrin, and their mRNA levels were considerably higher in a resistant population compared to susceptible strains. Recombinant CpGSTd3 was found to have the highest metabolic rate for lambda-cyhalothrin, supporting the role of GSTs in sequestration of the insecticide.

Summary: This study identified GSTs genes from the rice stem borer Chilo suppressalis and analyzed their classification, sequence characteristics, and genome structures. The research found that specific GSTs genes were significantly upregulated in C. suppressalis after treatment with abamectin, indicating their association with abamectin detoxification. These findings provide valuable information on the role of GSTs in abamectin detoxification in C. suppressalis and other insects.

High frequency of ryanodine receptor and cytochrome P450 CYP9A186 mutations in insecticide-resistant field populations of Spodoptera exigua from China

Haiyuan Teng et al.

Summary: The beet armyworm, a global agricultural pest, has developed resistance to insecticides such as chlorantraniliprole and emamectin benzoate. Mutations in specific genes, such as SeRyR and CYP9A186, have been found to contribute to resistance. However, the resistance mechanisms may vary for different insecticides. Therefore, the use of insecticides with different modes of action and other integrated pest management strategies are necessary to effectively control the beet armyworm in China.

Effect of RNAi Targeting SeGrx1 on the Cytotoxicity and Insecticide Susceptibility of Camptothecin in Spodoptera exigua

Fulai Yang et al.

Summary: This study investigated the effects of RNAi targeting SeGrx1 on the cytotoxicity and insecticide susceptibility of camptothecin (CPT) in Spodoptera exigua. The results showed that silencing SeGrx1 significantly increased the cytotoxicity of CPT to S. exigua cells, and the mortality of S. exigua larvae also increased. This suggests that SeGrx1 plays an important role in protecting cells from CPT-induced cytotoxicity and its silencing increases the sensitivity of S. exigua larvae to CPT.

Summary: Overall, the study found that CPT has high toxicity against Spodoptera litura larvae, causing growth stagnation and high mortality. Various energy-related metabolites were significantly affected by 0.5% CPT, while MAT showed low toxicity and minimal impact on hemolymph metabolites. Enzymatic and gene expression tests further supported the stronger toxicological effect of CPT compared to MAT against S. litura.

Comparative analysis of the detoxification gene inventory of four major Spodoptera pest species in response to xenobiotics

Dries Amezian et al.

Summary: The genus Spodoptera is known for being polyphagous and destructive agricultural pests, with detoxification genes playing a crucial role in overcoming plant defense mechanisms. Despite a significant amount of expression data produced in the past decade for Spodoptera larvae in response to xenobiotics, the scattered information makes it difficult to clearly identify candidate genes involved in host plant adaptation and insecticide resistance. This review highlights the importance of detoxification genes in the genus and reveals a lack of genetic and biochemical information in some species, suggesting a need for further research in this area.

Summary: The study identified rubitecan, a synthetic derivative of camptothecin, as a compound that reduced larval motility of Aedes aegypti mosquitoes. Further experiments showed that camptothecin and its derivatives also affected adult mosquitoes, increasing lethality and blocking egg laying at certain concentrations. These compounds are inhibitors of topoisomerase I and have potential for dual targeting of mosquito vectors and arboviruses like Zika virus.

Genome mapping coupled with CRISPR gene editing reveals a P450 gene confers avermectin resistance in the beet armyworm

Yayun Zuo et al.

Summary: Insecticide resistance is a global challenge for agricultural production, and the rapid identification of resistance genes is crucial for effective monitoring and management of resistant insect pests. This study reports a high-quality chromosome-level assembly of the beet armyworm genome and identifies the locus of avermectin resistance using BSA, with further confirmation of the involvement of CYP9A186 gene through gene editing. The natural substitution in the CYP9A186 protein results in enhanced metabolism of avermectins, highlighting the importance of coupling gene editing with BSA for identifying metabolic resistance genes.
