Summary: Cell-free protein synthesis (CFPS) is a versatile biotechnology platform with applications in clinical diagnostics, therapeutics production, and research. However, CFPS systems are time-consuming and expensive. This work presents a new CFPS system that reduces reagent cost and preparation time by eliminating the need for separately purified DNA templates. This system shows potential for cost reduction and use in low-resource settings.

Summary: This study leverages the advantages of CFPS combined with kinetic analysis, autoradiography, and western blot to elucidate the molecular mechanism of a biosensor for nuclear hormone receptor ligands. Enzyme kinetics simulations are used to optimize the biosensor assay, resulting in a longer readable window and stronger sensor signal. Creation of an on-demand thyroid hormone biosensor with observable color-change readout and the promise to improve methods for detecting natural hormones, therapeutic receptor modulators, and EDCs are significant outcomes of this approach.

Summary: Bioavailable glutamine (Gln) is essential for metabolism, intestinal health, immune function, and cell signaling. Routine measurement of serum Gln concentrations can aid in the improved diagnosis and treatment of severe infections, anorexia nervosa, chronic kidney disease, diabetes, and cancer, yet current methods may underutilize patient Gln levels. The use of Cell-free protein synthesis (CFPS) as a sensing platform for detecting Gln concentrations in human samples shows promising clinical applications and may lead to transformative point-of-care diagnostics and treatment regimens for various diseases.

Decentralizing Cell-Free RNA Sensing With the Use of Low-Cost Cell Extracts

Summary: Cell-free gene expression systems combined with programmable toehold switch-based RNA sensors offer a promising platform for field-deployed biosensing. However, the high commercial cost and cold-chain shipping requirements of current systems hinder implementation in resource-limited regions. By developing locally produced RNA toehold switch-based sensors using lysates, the cost can be reduced significantly while maintaining comparable performance to commercial systems. Implementing low-cost cell-free sensors could enhance biosensing capacity at the regional level and enable decentralized models for global surveillance of infectious diseases.
