Rapid automatic nucleic acid purification system based on gas-liquid immiscible phase

Summary: This study proposes a fast, highly automated, and low-consumption nucleic acid extraction method. By analyzing the process of magnetic beads crossing the gas-liquid interface, large magnetic bead packages were driven to cross the interface using a low magnetic field strength. This method provides a solution for high magnetic bead recovery rate in solid-phase extraction with a low-surfactant system based on a gas-liquid immiscible phase valve. The recovery rate of magnetic beads was improved to 90%-95%, and the carryover of reagents was below 1%.

From Biowaste to Lab-Bench: Low-Cost Magnetic Iron Oxide Nanoparticles for RNA Extraction and SARS-CoV-2 Diagnostics

Summary: The gold standard for COVID-19 diagnosis is based on RT-PCR using centralized facilities and commercial viral RNA extraction kits. Due to shortages at the beginning of the pandemic, alternative low-cost magnetic nanoparticles (MNPs) were developed from biowaste for viral RNA extraction. These MNPs, with their unique nanoplatelet shape and high surface area, showed comparable performance to commercial magnetic beads in high throughput PCR analysis and could significantly reduce the cost of nucleic acid extraction.

Pseudomonas aeruginosa detection based on droplets incubation using an integrated microfluidic chip, laser spectroscopy, and machine learning

Summary: This study proposes a novel pathogen detection system based on cultivating microdroplets and acquiring scattered light signals using a microfluidic device. The microdroplets are initially generated and incubated to cultivate bacteria. The features obtained are trained using machine learning classifiers, and the k-nearest neighbors algorithm demonstrates superior classification performance with 95.6% accuracy for identifying P. aeruginosa.

High-throughput quantitative detection of triple-negative breast cancer-associated expressed miRNAs by rolling circle amplification on fluorescence-encoded microspheres

Summary: Compared with other types of breast cancer, triple-negative breast cancer (TNBC) has a high degree of malignancy and poor prognosis. Early diagnosis of TNBC through specific miRNA expression profiles can reduce its mortality. In this study, we developed a high-throughput, high-sensitivity method for quantitatively detecting multiple sets of TNBC-associated miRNAs using rolling circle amplification on fluorescence-encoded microspheres. The method showed extensive linear dynamic range, high sensitivity, reasonable specificity, and good recovery and reproducibility in serum.

Highly sensitive smartphone-based detection of Listeria monocytogenes using SYTO9

Current LM detection methods are time-consuming and have low sensitivity. The method was simple, sensitive, and rapid, providing a new approach for the rapid detection of LM and other foodborne bacterial pathogens.

Ammonium-based ionic liquids (ILs) play an important role in ABS, and their structures have an influence on phase formation. By carefully selecting the components of ABS, a biocompatible system with controlled pH can be obtained for the partitioning of biomolecules. Experimental results showed that ABS containing benzyltributylammoniumchloride ([BTBA]Cl) IL exhibited high efficiency in DNA partitioning.

Magnetic Nylon 6 Nanocomposites for the Microextraction of Nucleic Acids from Biological Samples
