An insight into sex-specific neurotoxicity and molecular mechanisms of DEHP: A critical review

Yiyun Liu et al.

Summary: DEHP is widely used in various products as an additive to give plastics flexibility. However, its unstable combination with polymer leads to migration into the environment and subsequent human exposure. Studies have shown that DEHP exposure is associated with neurodevelopmental abnormalities and neurological diseases. Mechanisms such as oxidative damage, apoptosis, and signal transduction disorder contribute to neurobehavioral abnormalities and nerve injury caused by DEHP. Exposure to DEHP during critical developmental periods increases the risk of neurobehavioral abnormalities, depression, and autism spectrum disorders, with sex-specific effects that can persist into adulthood and have inter-generational effects. Future investigations should focus on population-scale and model organism studies to produce consistent and convincing results, while emphasizing the importance of mechanism research to better understand the environmental and human health risks of DEHP exposure.

Cumulative health risk in children and adolescents exposed to bis(2-ethylhexyl) phthalate (DEHP)

Xiangyun Gao et al.

Summary: DEHP contamination poses potential health risks for children and adolescents, primarily through dietary and nondietary exposure. Analysis of monitoring data reveals that the general population may face high risks of overexposure to DEHP, particularly schoolchildren aged 6 to 9.

Di(2-ethylhexyl) phthalate disturbs cholesterol metabolism through oxidative stress in rat liver

Gang Li et al.

Summary: This study investigated the effect of DEHP on hepatic cholesterol metabolism and revealed the mechanisms underlying DEHP-induced hepatotoxicity. The results showed that DEHP increased levels of total cholesterol, high-density lipoprotein, and alanine transaminase in the serum, and altered oxidative stress response and cholesterol metabolism pathway in the liver.

DEHP induce cholesterol imbalance via disturbing bile acid metabolism by altering the composition of gut microbiota in rats

Zhen Yu et al.

Summary: DEHP exposure induced cholesterol imbalance in rats, disrupted gut microbiota diversity, increased the ratio of Firmicutes to Bacteroidetes, and activated FXR in the gut, subsequently suppressing bile acid synthesis in the liver.

The impact of Di-(2-ethylhexyl) Phthalate and Mono(2-ethylhexyl) Phthalate in placental development, function, and pathophysiology

Luis Daniel Martinez-Razo et al.

Summary: DEHP, considered an endocrine disruptor chemical (EDC), is widely used in the plastic industry and can have negative effects on placental development and function, potentially impacting fetal growth. Research indicates that DEHP and MEHP can impair key processes in placental development and affect various placental functions. Clinical evidence also supports the connection between DEHP exposure and adverse pregnancy outcomes and pathologies.

Metabolic, reproductive and thyroid effects of bis(2-ethylhexyl) phthalate (DEHP) orally administered to male and female juvenile rats at dose levels derived from children biomonitoring study

Roberta Tassinari et al.

Summary: DEHP, an endocrine disrupter, was studied for its juvenile toxicity, revealing liver and metabolic system as main targets in both sexes, with thyroid issues in female rats and reproductive system effects in males. The study concluded that DEHP exposure during peri-pubertal period can induce sex-specific imbalances, utilizing a juvenile animal model for reliable risk assessment targeted towards children.

Lindong Han et al.

Summary: DEHP exposure during prepubertal stage can lead to testicular injury, characterized by reduced testicular volume, abnormal hormone levels, and structural damage. Transcriptome profiling identified differential gene expression related to key signaling pathways in response to DEHP exposure.
