T7 peptide-decorated exosome-based nanocarrier system for delivery of Galectin-9 siRNA to stimulate macrophage repolarization in glioblastoma

Chenguang Li et al.

Summary: This study evaluated the potential effect of T7 peptide-decorated exosome-loaded Galectin-9 siRNA (T7-Exo/siGalectin-9) in the M1 polarization of macrophages and immunosuppression of glioblastoma (GBM). The results showed that T7-Exo/siGalectin-9 effectively delivered siGalectin-9 to GBM cells and promoted macrophage polarization through the TLR7-IRF5 pathway, enhancing the anti-tumor effect of CD8+ T cells and suppressing inflammatory responses.

Exploration and functionalization of M1-macrophage extracellular vesicles for effective accumulation in glioblastoma and strong synergistic therapeutic effects

Xiaojun Wang et al.

Summary: The researchers successfully developed M1-like macrophage-derived extracellular vesicles (M1EVs) for the treatment of glioblastoma multiforme (GBM). These M1EVs achieved synergistic immunomodulation, chemiexcited photodynamic therapy (CDT), and hypoxia-activated chemotherapy in GBM, leading to a potent therapeutic effect. The results demonstrated the excellent efficacy of CCA-M1EVs against GBM in cell-derived and patient-derived xenograft models.

A Virus-Mimicking Nucleic Acid Nanogel Reprograms Microglia and Macrophages for Glioblastoma Therapy

Xihui Gao et al.

Summary: Immunotherapy is a promising approach for treating cancers, but its effectiveness in glioblastoma treatment is limited due to the immunosuppressive tumor microenvironment. A virus-mimicking membrane-coated nucleic acid nanogel, Vir-Gel, has been developed to reprogram microglia and macrophages from a pro-invasive M2 phenotype to an anti-tumor M1 phenotype. This approach significantly enhances the targetability and cell uptake efficiency of therapeutic miRNA, demonstrating improved tumor inhibition efficacy.

miR-21 and miR-146a: The microRNAs of inflammaging and age-related diseases

Fabiola Olivieri et al.

Summary: Inflammaging is a key concept in understanding how aging contributes to the development of age-related diseases, with a focus on the role of miRNAs in regulating inflammatory pathways. The assessment of individual genetic make-up alone is insufficient to depict the trajectories of age-related inflammation, and studies have shown that circulating inflammamiRs could be used to measure the degree of inflammaging.

Exosomes and biomimetic nanovesicles-mediated anti-glioblastoma therapy: A head-to-head comparison

Jun-Yong Wu et al.

Summary: The study compared Exos and BNVs for brain tumor drug delivery, finding that BNVs derived from brain-derived endothelial cells are a promising alternative with higher yield and similar drug-loading capacity to natural exosomes. Despite different mechanisms, BNV/DOX showed similar therapeutic effects to Exo/DOX in inhibiting the progression of glioblastoma, with minimal systemic toxicity. The findings suggest that autologous BNVs are an effective alternative to Exos for brain tumor nanomedicine.

Single-Cell Transcriptomics of Glioblastoma Reveals a Unique Tumor Microenvironment and Potential Immunotherapeutic Target Against Tumor-Associated Macrophage

Xiaoteng Cui et al.

Summary: This study analyzed the activation status and transcriptional signatures of tumor-associated macrophages (TAMs) in GBM at a single-cell level. Different TAM compositions were identified in each GBM subtype, with SPI1 identified as a crucial regulator for TAM maturation and polarization. Targeting SPI1 in immunotherapy could enhance the treatment effect of conventional chemotherapy drugs in GBM.

BSA-MnO2-SAL multifunctional nanoparticle-mediated M1 macrophages polarization for glioblastoma therapy

Fuming Liang et al.

Summary: Glioblastoma is a type of brain tumor with a high fatality rate, difficult to treat due to the presence of the blood-brain barrier and the dependence of tumor growth on the tumor microenvironment. Targeting M-2 macrophages and repolarizing them into M-1 macrophages are important strategies for treating glioblastoma. The nanoparticle BMS, designed to respond to acidic and H2O2-rich microenvironments, shows promising results in inhibiting glioblastoma growth and improving immunosuppressive microenvironments.
