Telomerase deficiency and dysfunctional telomeres in the lung tumor microenvironment impair tumor progression in NSCLC mouse models and patient-derived xenografts

Sergio Pineiro-Hermida et al.

Summary: Non-small cell lung cancer (NSCLC) is a leading cause of cancer death, and tumor progression is influenced by the interaction between cancer cells and the tumor microenvironment. Increased copy number and mRNA expression of TERT, a catalytic subunit of telomerase, is associated with decreased survival in NSCLC patients. Targeting telomeres may be an effective therapeutic approach in NSCLC treatment.

Smoking-associated upregulation of CBX3 suppresses ARHGAP24 expression to activate Rac1 signaling and promote tumor progression in lung adenocarcinoma

Xin Jin et al.

Summary: The study identified that smoking-induced upregulation of CBX3 promotes lung adenocarcinoma progression by activating the ARHGAP24/Rac1 pathway, suggesting the CBX3/ARHGAP24/Rac1 axis as a potential therapeutic target for smoking-induced LUAD.

CBX3 accelerates the malignant progression of glioblastoma multiforme by stabilizing EGFR expression

Wen Peng et al.

Summary: CBX3, also known as HP1 gamma, is upregulated in glioblastoma multiforme (GBM) and promotes cell proliferation, invasion, and tumorigenesis. It regulates the malignant progression of GBM through an EGFR-dependent mechanism. CBX3 directly suppresses the transcriptional levels of PARK2 and STUB1, reducing the ubiquitination of EGFR.

Proteogenomic and metabolomic characterization of human glioblastoma

Liang-Bo Wang et al.

Summary: Understanding the molecular pathogenesis of glioblastoma is crucial for improving diagnosis and treatment. Integrated analysis of genomic, proteomic, and metabolomic data on treatment-naive glioblastomas has identified key phosphorylation events and potential targets for specific tumor alterations. Immune subtypes with distinct immune cell types were discovered, and specific histone acetylation patterns were correlated with these subtypes, providing insights for stratification of patients for more effective treatment.

Higher order genetic interactions switch cancer genes from two-hit to one-hit drivers

Solip Park et al.

Summary: The classic two-hit model suggests that both alleles of a tumor suppressor gene must be inactivated to cause cancer. However, research shows that many cancer genes can actually switch between acting as one-hit or two-hit drivers, depending on the context and other gene mutations. This indicates that the correct genetic model for a gene depends on other mutations in the genome, with a second hit in the same gene or an alteration in a different gene in the same pathway sometimes representing alternative evolutionary paths to cancer.

Rozita Bagheri-Yarmand et al.

Summary: This study identified a novel RAC1 (P34R) mutation acquired during dabrafenib treatment, as well as revealed that an increase in RAC1 copy numbers and polyploidy of chromosome 7 may contribute to drug resistance.
