What makes Yarrowia lipolytica well suited for industry?

Summary: Yarrowia lipolytica is a promising candidate for industrial bioproduction of chemicals, fuels, foods, and pharmaceuticals, thanks to its natural and engineered traits. In recent years, extensive research has been conducted to assess its potential, improve its features, and scale up its processes. The increasing number of publications and patents related to Y. lipolytica demonstrates its importance and potential in the field. Despite the remaining challenges, it is expected that Y. lipolytica will become one of the preferred choices for industrial applications in the near future.

Summary: In this paper, the authors propose a neural network-based architecture called DeepGuide, which learns from genome-wide CRISPR activity profiles to accurately design high activity sgRNAs. The experimental validation confirms the effectiveness of DeepGuide in predicting high activity sgRNAs in the oleaginous yeast Yarrowia lipolytica.

YALIcloneNHEJ: An Efficient Modular Cloning Toolkit for NHEJ Integration of Multigene Pathway and Terpenoid Production in Yarrowia lipolytica

Summary: In this study, a Golden Gate modular cloning system (YALIcloneNHEJ) was established to develop a robust DNA assembly platform in Yarrowia lipolytica. By optimizing key factors, the assembly efficiency of multiple fragments reached up to 90%. The system was successfully applied for the overproduction of (-)-alpha-bisabolol, achieving the highest titer reported in yeast to date. This study expands the toolbox of metabolic engineering and enables efficient production of various terpenoids.

Screening neutral sites for metabolic engineering of methylotrophic yeast Ogataea polymorpha

Summary: In this study, genomic neutral sites for gene integration were screened in O. polymorpha via CRISPR-Cas9 technique. 17 potential neutral sites were identified, with fatty alcohol synthetic pathway genes successfully integrated into two of them, resulting in the production of 4.5 mg/L fatty alcohols. This method of neutral sites screening shows promise for streamline metabolic engineering in O. polymorpha and potentially other eukaryotes as well.

CRISPR-mediated multigene integration enables Shikimate pathway refactoring for enhanced 2-phenylethanol biosynthesis in Kluyveromyces marxianus

Summary: The study focuses on engineering the Shikimate and Ehrlich pathways in yeast Kluyveromyces marxianus for the biosynthesis of 2-phenylethanol (2-PE). A CRISPR-mediated multigene integration system was developed to integrate multiple gene expression cassettes efficiently, leading to an increase in 2-PE production. The best strain achieved 1943 mg/L 2-PE after 120 h fed-batch operation in shake flask cultures, demonstrating the potential of this approach for sustainable and economical production.

Expansion of EasyClone-MarkerFree toolkit for Saccharomyces cerevisiae genome with new integration sites

Summary: This study expanded the EasyClone-MarkerFree vector toolkit for baker's yeast S. cerevisiae by adding eight additional intergenic integration sites. The integration efficiency at the new sites was above 80%, with similar or higher expression levels of green fluorescent protein compared to the original sites. Integration at the new locations did not affect cellular growth.

A Yarrowia lipolytica Strain Engineered for Pyomelanin Production

Summary: In this study, the yeast Yarrowia lipolytica was engineered to overproduce pyomelanin by optimizing a chassis strain to increase homogentisic acid synthesis. The overexpression of the 4HPPD gene led to significantly higher pyomelanin yields, with the pathway being strongly induced by phenylalanine supplementation.

Hybrid promoter engineering strategies in Yarrowia lipolytica: isoamyl alcohol production as a test study

Summary: This study systematically detected and characterized common promoter elements in Yarrowia lipolytica, and constructed an artificial hybrid promoter library covering a wide range of promoter strength. The hybrid promoter strength can be fine-tuned by promoter elements to optimize expression in Y. lipolytica. This promoter engineering strategy could be extended to other non-model fungi for strain improvement.
