New Fully Automated Preparation of High Apparent Molar Activity 68Ga-FAPI-46 on a Trasis AiO Platform

Summary: A large number of applications for FAPI-based PET agents have been evaluated, including in cancer and non-malignant diseases. Ga-68-FAPI-46 showed high specificity and affinity for FAP-expressing cells, with fast accumulation in tumor lesions/injuries and quick body clearance. An automated synthesis for the production of Ga-68-FAPI-46 on a Trasis AiO platform was developed, resulting in high radiochemical purity and molar activity. The product's in vitro and in vivo properties were assessed on glioblastoma cells and mouse model.

Automated Synthesis of 68Ga-Labeled DOTA-MGS8 and Preclinical Characterization of Cholecystokinin-2 Receptor Targeting

Summary: This study presents a new minigastrin analog, DOTA-MGS8, targeting the cholecystokinin-2 receptor (CCK2R). Ga-68-labeled DOTA-MGS8 was successfully synthesized using an automated synthesis module, and preclinical studies demonstrated its high receptor-specific cell internalization and favorable biodistribution profile. This suggests that stabilized MG analogs like DOTA-MGS8 have the potential for high-sensitivity positron emission tomography in patients with CCK2R-related malignancies.

Automated synthesis of [68Ga]Ga-FAPI-46 without pre-purification of the generator eluate on three common synthesis modules and two generator types

Summary: The study aimed to develop and evaluate a GMP-compliant automatic synthesis of [Ga-68]Ga-FAPI-46 using different Ge-68/Ga-68 generators and synthesis systems. Regardless of the generator or synthesis module used, the synthesis of [Ga-68]Ga-FAPI-46 could be completed in 12 minutes with over 99.5% chemical purity and a radiochemical yield of around 92.5% (decay corrected). All quality control parameters met the requirements of the European Pharmacopoeia.

Safety of Fibroblast Activation Protein-Targeted Radionuclide Therapy by a Low-Dose Dosimetric Approach Using 177Lu-FAPI04

Summary: This study aimed to estimate radiation-absorbed doses in normal organs and tumor tissue using low-dose Lu-177-FAPI04 dosimetry in metastatic cancer patients. Results showed low absorbed doses in risk organs, allowing for low tumor absorbed doses with higher administered doses. Further research on optimizing therapeutic efficacy and individualized dosimetric approach is needed.
