NIR Spectroscopy as an Online PAT Tool for a Narrow Therapeutic Index Drug: Toward a Platform Approach Across Lab and Pilot Scales for Development of a Powder Blending Monitoring Method and Endpoint Determination

Summary: An online NIR spectroscopy platform system was used to monitor real-time powder blending and determine blend endpoint in a phenytoin sodium formulation. The study investigated multivariate data acquisition, model development, and model scale-up from lab to manufacturing using robust experimental design and multiple sensors. Different algorithms for blend endpoint determination were explored to understand their impact on predicted blend endpoint. The results showed that algorithms sensitive to deviation from target concentration offered the simplest interpretation and consistent trends, while algorithms sensitive to global homogeneity yielded longer mixing time. Qualitative algorithms using PCA of spectral data predicted the shortest mixing time for blending endpoint. The hybrid approach of combining NIR data from different scales presented several advantages, simplifying the model building process and reducing costs compared to solely using data from commercial scale.
