Target enzymes in oxaliplatin-induced peripheral neuropathy in Swiss mice: A new acetylcholinesterase inhibitor as therapeutic strategy

Ketlyn P. da Motta et al.

Summary: In this study, it was found that the acetylcholinesterase inhibitor MTDZ has antinociceptive effects and can reduce the comorbidities associated with oxaliplatin-induced peripheral neuropathy. MTDZ reversed the hypersensitivity, cognitive impairment, and anxious-like behavior caused by oxaliplatin exposure. The results suggest that MTDZ is a promising molecule for the treatment of oxaliplatin-induced neurotoxicity.

Genistein reverses the effect of 17β-estradiol on exacerbating experimental occlusal interference-induced chronic masseter hyperalgesia in ovariectomised rats

Yun Liu et al.

Summary: This study found that female rats exhibited greater sensitivity to masseter hyperalgesia induced by occlusal interference compared to males, with the lowest sensitivity seen during the pro-oestrus phase. Genistein may counteract the enhancement of chronic masseter hyperalgesia by E2 through regulating TRPV1 expression in the trigeminal ganglion.

Glutamatergic Neurons in the Amygdala Are Involved in Paclitaxel-Induced Pain and Anxiety

Jiaxin Liu et al.

Summary: This study aims to elucidate the role of Ca2+/calmodulin-dependent protein kinase II (CaMKII)-positive glutamatergic neurons in the amygdala in paclitaxel-induced pain and negative affective symptoms. Selective inhibition of CaMKII neurons in the amygdala can alleviate pain and anxiety behaviors.

7-Chloro-4-(Phenylselanyl) Quinoline Is a Novel Multitarget Therapy to Combat Peripheral Neuropathy and Comorbidities Induced by Paclitaxel in Mice

Jaini J. Paltian et al.

Summary: Paclitaxel-induced peripheral neuropathy (PIPN) is a common and complex painful condition associated with paclitaxel (PTX) exposure, leading to decreased quality of life, anxiety, and cognitive impairment. However, there are limited treatment options available. In this study, the therapeutic potential of 4-PSQ was assessed for reversing PIPN and its comorbidities. The results showed that 4-PSQ reduced PTX-induced hypersensitivity, anxiety, and cognitive impairment, possibly through modulation of oxidative stress, neuroinflammation, and calcium homeostasis.

Mechanistic pathways of fibromyalgia induced by intermittent cold stress in mice is sex-dependently

Carolina C. Martins et al.

Summary: Fibromyalgia is a complex disease with unknown pathophysiology, making it challenging to develop appropriate treatments. This study examined the involvement of serotonergic receptors, the NMDA/NO/cGMP pathway, and oxidative stress in an animal model of fibromyalgia. The findings suggest that these factors may contribute to nociceptive behaviors induced by cold stress exposure, and the effects appear to be sex-dependent.

Current opinion on the pharmacogenomics of paclitaxel-induced toxicity

Zeina N. Al-Mahayri et al.

Summary: Paclitaxel is a commonly used chemotherapeutic agent with notable efficacy in various types of cancer, however it is associated with toxicities such as neurotoxicity. Studies have explored mechanisms and risk factors of paclitaxel-induced toxicities, with promising candidates found in genes related to neuro-sensitivity for future research. Further investigation into toxicity development mechanisms and personal vulnerability factors is needed to enhance understanding and management of paclitaxel-associated toxicities.
