Cell-in-cell structure mediates in-cell killing suppressed by CD44

Summary: Penetration of immune cells into tumor cells through cell-in-cell (CIC) formation leads to in-cell killing of the host tumor cells, showing superior efficacy compared to canonical kiss-killing from outside. CD44 on tumor cells functions as a negative regulator of in-cell killing by inhibiting CIC formation. Antibody-mediated blockade of CD44 signaling enhances the suppressive effects of NK cells on tumor growth.

Summary: This study found that colorectal tumor hybrid cells (THCs) generated from in vitro co-culture of colorectal cancer stem cells and human monocytes showed enhanced migratory, proliferative, and immune evasion abilities compared to their parental cells. The number of THCs was found to be associated with prognosis and the spread of metastasis in colorectal cancer patients, indicating that the number of THCs could serve as a novel biomarker for metastasis prediction.

Direct cell-to-cell transfer in stressed tumor microenvironment aggravates tumorigenic or metastatic potential in pancreatic cancer

Summary: In this study, an in vitro tumor microenvironment mimic system was established and it was found that the direct transfer of biomolecules between cells increased under stress conditions. Further experiments showed that this transfer promoted the growth, invasiveness, and metabolic reprogramming of pancreatic cancer cells. Moreover, CD44 and MCT1 were identified as potential targets for preventing recurrence of pancreatic cancer.
