O-GlcNAcylation of MORC2 at threonine 556 by OGT couples TGF-beta signaling to breast cancer progression

Ying-Ying Liu et al.

Summary: This study reveals the importance of MORC2 O-GlcNAcylation in breast cancer progression. MORC2 is O-GlcNAcylated by OGT at threonine 556, and this modification is required for its role in breast cancer cell migration and invasion. TGF-beta 1 induces MORC2 O-GlcNAcylation through enhancing the stability of GFAT, and O-GlcNAcylated MORC2 is necessary for the transcriptional activation of TGF-beta 1 target genes CTGF and SNAIL. These findings provide evidence for targeting MORC2-dependent breast cancer through blocking its O-GlcNAcylation.

DHODH-mediated ferroptosis defence is a targetable vulnerability in cancer

Chao Mao et al.

Summary: The study reveals the mechanism of ferroptosis induced by inhibiting GPX4 in cancer cells, as well as the potential pathways to enhance or inhibit ferroptosis by intervening in relevant metabolic pathways. Therapeutic strategies targeting different levels of GPX4 expression could be a promising new approach for cancer treatment.

HIF-2α activation potentiates oxidative cell death in colorectal cancers by increasing cellular iron

Rashi Singhal et al.

Summary: This study reveals the essential role of HIF-2α in colorectal cancer (CRC) progression, demonstrating its regulation of cell death via modulation of cellular iron and oxidation. Inhibitors targeting HIF-2α can enhance cancer cells' susceptibility to oxidative cell death.
