Asperuloside Prevents Peri-Implantitis via Suppression of NF-κB and ERK1/2 on Rats

Xinge Wang et al.

Summary: Peri-implantitis is a condition characterized by inflammation and bone resorption, leading to implant failure. Asperuloside (ASP), an iridoid glycoside, has shown significant anti-inflammatory properties and the potential to attenuate peri-implantitis bone resorption. This study found that ASP could inhibit osteoclast formation, decrease pro-inflammatory cytokine levels, and mitigate bone resorption in peri-implantitis.

Finely-Tuned Calcium Oscillations in Osteoclast Differentiation and Bone Resorption

Hiroyuki Okada et al.

Summary: Calcium oscillations play a crucial role in osteoclast differentiation and function, with various modifiers involved in fine-tuning this process. In addition to traditional factors like M-CSF and RANKL, immunoreceptor-related adaptors' costimulatory signaling is important for calcium oscillation generation. Certain calcium channels and transporters are essential for calcium oscillations in osteoclasts, alongside organelles like lysosomes, mitochondria, and the ER storing intracellular calcium.

Role of K+ and Ca2+-Permeable Channels in Osteoblast Functions

Hiroaki Kito et al.

Summary: Osteoblasts are crucial in bone modeling and remodeling, with their differentiation being regulated by various signaling pathways. Proper bone homeostasis depends on the differentiation and function of osteoblast lineage cells. Ca2+ signaling controls many processes in osteoblast lineage cells, and K+ channel activation indirectly influences Ca2+ signaling.

Yukmijihwang-Tang Suppresses Receptor Activator of Nuclear Factor Kappa-B Ligand (RANKL)-Induced Osteoclast Differentiation and Prevents Ovariectomy (OVX)-Mediated Bone Loss

Sang-Yong Han et al.

Summary: Yukmijihwang-tang (YJ) was found to reduce osteoclast differentiation and bone loss in rats by regulating the RANKL/OPG ratio, inhibiting NFATc1 and c-Fos activation, and suppressing early signaling pathways. These results suggest the potential therapeutic application of YJ for bone diseases like osteoporosis.

Inhibitory Effect of a Rosmarinic Acid-Enriched Fraction Prepared from Nga-Mon (Perilla frutescens) Seed Meal on Osteoclastogenesis through the RANK Signaling Pathway

Kanokkarn Phromnoi et al.

Summary: The study aimed to investigate the antioxidant and anti-inflammatory effects of various perilla seed meal fractions and their impact on osteoporosis. The ethyl acetate fraction of seed meal showed the highest antioxidant and anti-inflammatory activities, with a high content of rosmarinic acid. It was found that this fraction effectively suppressed RANKL-induced osteoclast formation by downregulating NF-kappa B, AP-1 activation, and NFATc1 expression, and inhibited ROS production as well. These results suggest that the ethyl acetate fraction rich in rosmarinic acid has potential for osteoclastogenic protection and health promotion.
