Mindin (SPON2) Is Essential for Cutaneous Fibrogenesis in a Mouse Model of Systemic Sclerosis

Isha Rana et al.

Summary: It was discovered that the transcription factor SNAI1 is overexpressed in the epidermis of patients with systemic sclerosis, and a transgenic mouse model mimicking this expression pattern can induce many clinical features of the human disease. The study found that Mindin protein plays a critical role in fibrogenesis by inducing early inflammatory cytokine production and collagen secretion in resident dermal fibroblasts. Targeting Mindin protein holds promise as an effective therapy for fibrosis.

RSPO2 defines a distinct undifferentiated progenitor in the tendon/ligament and suppresses ectopic ossification

Naohiro Tachibana et al.

Summary: This study identified that R-spondin 2 (RSPO2) is specifically expressed in a distinct Prg4(+) tendon stem/progenitor cell (TSPC) cluster and suppresses ectopic ossification via chondrogenic differentiation suppression. RSPO2 expression levels were lower in patients with ossification of the posterior longitudinal ligament compared to spondylosis patients, and RSPO2 protein suppressed chondrogenic differentiation in human ligament cells.

The fibrillin microfibril/elastic fibre network: A critical extracellular supramolecular scaffold to balance skin homoeostasis

Christin S. Adamo et al.

Summary: Supramolecular networks composed of fibrillins and associated ligands play a crucial role in balancing skin homoeostasis and directing remodeling, influencing skin elasticity and growth factor availability. Fibrillin microfibrils are essential for elastic fiber formation and contribute to skin pliability. Mutations in fibrillin genes can lead to a wide range of skin phenotypes, with FMF regulating growth factor signaling critical for skin growth and homoeostasis.

Collagen XII mediated cellular and extracellular mechanisms regulate establishment of tendon structure and function

Yayoi Izu et al.

Summary: Collagen XII plays dual roles in tendon development by influencing fibril packing, fiber assembly and stability, as well as regulating tenocyte organization and intercellular communication. Its deficiency results in altered cell shape, impaired cell-cell communication, and decreased tissue stiffness, indicating its crucial role in tendon structure and function. Additionally, collagen XII specifically affects tenocyte biosynthesis and provides feedback to regulate extracellular collagen I, highlighting its significance in tendon development and maintenance.

Mitochondrial respiratory chain function promotes extracellular matrix integrity in cartilage

Kristina Bubb et al.

Summary: This study utilized advanced technologies to investigate the significant impact of mitochondrial respiratory chain on ECM homeostasis in mature cartilage, revealing that dysfunction of the mitochondrial respiratory chain can lead to increased ECM deposition and stiffness. These findings highlight the crucial role of the mitochondrial respiratory chain in maintaining ECM integrity and mechanical stability in cartilage.
