Empagliflozin attenuates the renal tubular ferroptosis in diabetic kidney disease through AMPK/NRF2 pathway

QianYu Lu et al.

Summary: Renal tubular damage is a key factor in the development of diabetic kidney disease (DKD), with ferroptosis as the main pathological process. Empagliflozin has been suggested as a potential treatment for renal injury, but its effects on diabetic-related ferroptosis and underlying mechanisms are not fully understood. This study evaluated the influence of empagliflozin on renal injury using mouse models and cell lines, and found that it can decrease ferroptosis in DKD mice and high-glucose stimulated cells. The protective effects of empagliflozin are partially mediated by AMP-activated protein kinase (AMPK) and the NRF2 activation pathway. These findings offer important insights for novel treatment approaches for DKD.

Ferroptosis triggered by STAT1-IRF1-ACSL4 pathway was involved in radiation-induced intestinal injury

Peizhong Kong et al.

Summary: Radiation-induced intestinal injury (RIII) is a common complication of radiotherapy, seriously affecting patients' quality of life. This study found that ferroptosis plays a critical role in RIII, with increased levels of lipid peroxidation markers. In vivo experiments demonstrated that the ferroptosis inhibitor improved the survival rate and alleviated intestinal fibrosis in mice. It was also found that arachidonic acid enhanced ferroptosis in cultured intestinal epithelial cells and aggravated RIII. Mechanistic studies revealed that the level of ACSL4 protein increased after irradiation, regulated by the STAT1/IRF1 axis, while AMPK activation triggered by AA negatively regulated radiation-induced ferroptosis.

Epigenetic regulation of ferroptosis via ETS1/miR-23a-3p/ACSL4 axis mediates sorafenib resistance in human hepatocellular carcinoma

Yuanjun Lu et al.

Summary: Our study reveals that the ETS1/miR-23a-3p/ACSL4 axis contributes to sorafenib resistance in HCC by regulating ferroptosis. This suggests that miR-23a-3p could be a potential target for improving sorafenib responsiveness in HCC patients.

PGK1 contributes to tumorigenesis and sorafenib resistance of renal clear cell carcinoma via activating CXCR4/ERK signaling pathway and accelerating glycolysis

Yu He et al.

Summary: This study comprehensively explores the molecular mechanisms of PGK1 in the progression and sorafenib resistance of KIRC. The upregulation of PGK1 is associated with poor prognosis in KIRC patients. Furthermore, PGK1 promotes tumor growth and sorafenib resistance in KIRC through regulating glycolysis and the CXCR4/ERK signaling pathway.

Cardiac-specific overexpression of Ndufs1 ameliorates cardiac dysfunction after myocardial infarction by alleviating mitochondrial dysfunction and apoptosis

Bingchao Qi et al.

Summary: This study demonstrates that decreased expression of Ndufs1 is associated with heart failure after myocardial infarction. Cardiac-specific overexpression of Ndufs1 alleviates cardiac dysfunction and myocardial fibrosis. Ndufs1 overexpression also reduces ROS production and apoptosis, and improves mitochondrial respiratory function. Targeting Ndufs1 may be a potential therapeutic strategy for improving cardiac function in patients with heart failure after myocardial infarction.

Ferroptosis: molecular mechanisms and health implications

Daolin Tang et al.

Summary: Cell death can be executed through different subroutines, such as ferroptosis which is an iron-dependent form of non-apoptotic cell death. Ferroptosis can occur through two major pathways, driven by redox imbalance and abnormal expression of redox-active enzymes. The process is precisely regulated at multiple levels, with the transcription factor NFE2L2 playing a central role in anti-ferroptotic defense.
