Tendon Healing Response Is Dependent on Epithelial-Mesenchymal-Tendon Transition State of Amniotic Epithelial Stem Cells

Valentina Russo et al.

Summary: This research demonstrates the potential of amniotic epithelial stem cells (AECs) in promoting tendon regeneration. The study found that AECs can trans-differentiate into cell phenotypes with varying abilities to facilitate regeneration. The findings suggest that AECs with an epithelial phenotype have a greater impact on shifting the inflammatory response to a pro-regenerative response, while AECs with a tendon-like phenotype mainly affect the proliferative phase of regeneration. These results provide support for using AECs as a feasible and effective strategy for the treatment of acute tendinopathies.

In medio stat virtus: Insights into hybrid E/M phenotype attitudes

Angelo Canciello et al.

Summary: Epithelial-mesenchymal plasticity (EMP) refers to the ability of cells to interconvert between epithelial and mesenchymal phenotypes, generating hybrid intermediate cells with mixed E and M features. These hybrid cells play key roles in physiological and pathological processes, including stemness gene expression, differentiation, wound healing, immune suppression, and cancer cell characteristics such as metastasis and therapy resistance.

The MHC Class II Transactivator CIITA: Not (Quite) the Odd-One-Out Anymore among NLR Proteins

Jorge Alfonso Leon Machado et al.

Summary: CIITA is the master regulator of MHC class II gene expression, functioning as a non-DNA-binding activator through interactions with the MHC enhanceosome complex. In addition to its role in MHC-II gene regulation, CIITA also plays important roles in cancer, viral restriction, and inhibition of virus replication.

Cellular dynamics of EMT: lessons from live in vivo imaging of embryonic development

Jeffrey D. Amack

Summary: Epithelial-mesenchymal transition (EMT) is a process where cells lose polarity and gain invasive properties, used in both embryonic development and wound healing in adults, as well as exploited by cancer cells. Regulation of EMT involves rapid changes in protein expression in response to cell signaling and environmental cues. Live imaging in model organisms allows for insights into EMT mechanisms, highlighting the importance of cytoskeletal dynamics in this process.

Regulation of immune responses by the airway epithelial cell landscape

Richard J. Hewitt et al.

Summary: The community of cells lining our airways collaborates in maintaining immune homeostasis in the lung and protecting against pathogens and pollutants in the air. Single-cell RNA-sequencing reveals cellular heterogeneity in the airway wall and identifies novel cell populations with unique molecular signatures and functions in health and disease. This review discusses the evolving understanding of the airway epithelial landscape with a focus on interactions with the local neuronal and immune systems.
