Circulating tumor cell gene expression and plasma AR gene copy number as biomarkers for castration-resistant prostate cancer patients treated with cabazitaxel

Giorgia Gurioli et al.

Summary: This study evaluated the prognostic role of CTC gene expression in cabazitaxel-treated mCRPC patients and its association with plasma AR copy number. The results showed that CTC gene expression was significantly associated with overall survival and disease progression. AR-V7 was identified as an important prognostic biomarker, and patients with AR-V7 positivity had poorer outcomes with lower dose cabazitaxel treatment.

Summary: Each year, the American Cancer Society compiles data on cancer occurrence and outcomes in the United States. The latest data shows that breast and prostate cancer progress has stagnated, while lung cancer has shown improvements in survival rates. Lung cancer incidence for advanced disease has declined while localized-stage rates have increased, resulting in higher survival rates. Mortality patterns align with incidence trends, with lung cancer deaths declining rapidly, breast cancer deaths slowing, and prostate cancer deaths stabilizing.

Circ_0004087 interaction with SND1 promotes docetaxel resistance in prostate cancer by boosting the mitosis error correction mechanism

Liang Chen et al.

Summary: Our study revealed the interaction between circ_0004087 and the mitosis error correction mechanism, showing that circ_0004087 can stimulate the expression of BUB1 through SND1 and MYB, leading to enhanced mitotic accuracy of PCa cells. Furthermore, circ_0004087 was found to confer DTX resistance in PCa cells, suggesting its potential as a prognostic biomarker and therapeutic target in DTX-resistant PCa patients.

bub1 as a potential oncogene and a prognostic biomarker for neuroblastoma

Jingjing Song et al.

Summary: In this study, a potential tumor-promoting gene bub1 for neuroblastoma was identified and found to also serve as a prognostic biomarker. The bub1 gene was shown to promote cancer progression in neuroblastoma by regulating important downstream molecules related to apoptosis and epithelial-mesenchymal transition.

Comparative analysis of 1152 African-American and European-American men with prostate cancer identifies distinct genomic and immunological differences

Walter Rayford et al.

Summary: Racial disparities in prostate cancer were investigated in a multi-institutional retrospective analysis of African-American and European-American men who underwent radical prostatectomy. Differences were found in gene expression with African-American men showing higher expression of inflammation genes and lower expression of mismatch repair genes.

Hormonal Therapy for Prostate Cancer

Kunal Desai et al.

Summary: Huggins and Hodges demonstrated the therapeutic effect of gonadal testosterone deprivation in the 1940s, leading to the concept of prostate cancer being highly androgen-dependent. Since then, hormonal therapy has evolved from gonadal testosterone deprivation to inhibiting extragonadal androgens, to directly binding and inhibiting the androgen receptor. Today's standard of care for advanced prostate cancer includes gonadotropin-releasing hormone agonists and androgen receptor antagonists.

Androgen receptor splicing variant 7 (ARV7) inhibits docetaxel sensitivity by inactivating the spindle assembly checkpoint

Bingbing Yu et al.

Summary: The study revealed that ARV7 mediates DTX sensitivity by inactivating SAC and promoting mitotic slippage, leading to resistance. By regulating E2 enzyme UBE2C, ARV7 plays a role in promoting SAC inactivation and early degradation of cyclin B1. Combination treatment of DTX and a mitotic exit inhibitor shows a synergistic effect in high ARV7-expressing prostate cancer cells.

BUB1 drives the occurrence and development of bladder cancer by mediating the STAT3 signaling pathway

Ning Jiang et al.

Summary: The study revealed that increased expression of BUB1 in human bladder cancer was associated with the activation of STAT3 signaling. Inhibition of BUB1 kinase activity effectively suppressed BCa cell proliferation and cancer progression both in vitro and in vivo. The BUB1/STAT3 complex played a critical role in promoting the transcription of STAT3 target genes, highlighting the potential of BUB1 as a therapeutic target for bladder cancer treatment.

Aneuploidy renders cancer cells vulnerable to mitotic checkpoint inhibition

Yael Cohen-Sharir et al.

Summary: The study shows that aneuploid cancer cells are more sensitive to genetic perturbation of SAC components and their long-term proliferation is jeopardized. Furthermore, aneuploid cells exhibit aberrant spindle geometry and dynamics, which result in mitotic defects and less-fit karyotypes over time.

KIF4A knockdown suppresses ovarian cancer cell proliferation and induces apoptosis by downregulating BUB1 expression

Wumin Jin et al.

Summary: The study elucidated that KIF4A suppresses ovarian cancer progression by regulating BUB1, with KIF4A knockdown significantly inhibiting cell viability, colony formation, and migration, while inducing apoptosis. The results suggest the potential therapeutic value of KIF4A and BUB1 as targets for ovarian cancer treatment.
