CHIPing away the progression potential of CHIP: A new reality in the making

Zhuoer Xie et al.

Summary: In recent years, there has been an increased understanding of the epidemiology, clinical implications, and mechanistic aspects of clonal hematopoiesis of indeterminate potential (CHIP). However, interventions to prevent or delay the negative consequences of CHIP are still lacking. This review highlights the latest updates on CHIP research, including molecular mechanisms and clinical implications, with a focus on the evolving strategies for interventions in ongoing trials. Standardized and evidence-based recommendations and guidelines are urgently needed for the evaluation and management of individuals with clonal hematopoiesis. Patient-centric endpoints must also be defined for clinical trials to further develop effective preventive strategies and improve clinical outcomes.

Discovery of novel predisposing coding and noncoding variants in familial Hodgkin lymphoma

Jamie E. Flerlage et al.

Summary: Familial aggregation of HL has been shown to have a genetic component. In this study, whole genome sequencing was performed on individuals with and without HL from pedigrees with multiple affected relatives. A total of 44 HL-risk variants were identified, including both coding and noncoding variants. The study also identified several novel variants and observed segregation patterns in most pedigrees.

Pathology review identifies frequent misdiagnoses in recurrent classic Hodgkin lymphoma in a nationwide cohort: implications for clinical and epidemiological studies

Max V. Boot et al.

Summary: A study in the Netherlands re-evaluated the risk of developing non-Hodgkin lymphoma (NHL) after classic Hodgkin lymphoma (CHL) treatment and identified pathological features that resemble CHL. The study found that the risk of developing NHL after CHL treatment is lower than previously reported and highlighted the importance of accurate diagnosis and treatment.

Clonality assessment and detection of clonal diversity in classic Hodgkin lymphoma by next-generation sequencing of immunoglobulin gene rearrangements

Diede A. G. van Bladel et al.

Summary: The clonality analysis plays an important role in diagnosing classic Hodgkin lymphoma, especially in cases with atypical presentation or histology resembling T cell lymphoma and patients with recurrent disease. The development of a novel NGS-based assay has significantly improved the detection of clonal gene rearrangements in cHL cases, particularly in formalin-fixed paraffin-embedded tissue specimens, demonstrating increased sensitivity and performance compared to conventional methods.

Novel Approaches in Molecular Characterization of Classical Hodgkin Lymphoma

Diede A. G. van Bladel et al.

Summary: This review provides an overview of recent advances in genotyping the clonal and mutational landscape of classical Hodgkin lymphoma (cHL). The authors discuss the application of next-generation sequencing in the diagnosis and treatment of cHL, as well as its contribution to understanding the pathogenesis of cHL.

Classical Hodgkin lymphoma

Pauline Brice et al.

Summary: Classical Hodgkin lymphoma is a relatively common lymphoma that is generally considered highly curable with standard first-line chemotherapy and radiotherapy, although some patients may not be cured or may experience late toxic effects leading to premature death. Recent changes in the gold standard of chemotherapy, now often including immunotherapy in relapse settings, have further impacted treatment outcomes.

Molecular biology of Hodgkin lymphoma

Marc A. Weniger et al.

Summary: Classical Hodgkin lymphoma (cHL) has unique biological features such as the orchestration of a complex microenvironment by HRS cells, the major role of Epstein-Barr virus in lymphomagenesis, and the defining interplay of genetic lesions and cellular origin. These factors contribute to the constitutive activation of multiple signaling pathways in cHL.

Summary: The study identified specific mutations that promote progression in myeloid neoplasms patients and found that patients with progression have more genetic alterations. Furthermore, it emphasized the importance of both the quantity and order of genetic alterations in disease evolution.

Next-Generation Sequencing-Based Clonality Assessment of Ig Gene Rearrangements A Multicenter Validation Study by EuroClonality-NGS

Michiel van den Brand et al.

Summary: NGS-based IG clonality analysis shows high interlaboratory concordance and concordance with conventional analysis, with a higher ability to detect clonal rearrangements. NGS and conventional analysis detect clonality in 96% and 95% of B-cell neoplasms, respectively, with the majority of reactive cases being polyclonal.

Dissecting Clonal Hematopoiesis in Tissues of Patients with Classic Hodgkin Lymphoma

Alessandra Venanzi et al.

Summary: The study found that clonal hematopoiesis could be present in the tissue of classic Hodgkin lymphoma (cHL), give rise to the tumor clone, and spread to large parts of its microenvironment. Even when massive, clonal hematopoiesis does not always lead to the neoplastic clone of multiple myeloid and lymphoid neoplasms occurring in the same patient.
