Catechin potentiates the antifungal effect of miconazole in Candida glabrata

Summary: The increasing number of drug-resistant Candida strains causing invasive fungal infections poses a major challenge to the development of new antifungal strategies. Researchers have turned their attention to natural products, such as catechins, as potential antifungals and in combination therapies. In this study, the combination of catechin and antifungal azoles was found to inhibit the growth of Candida glabrata isolates, including azole-resistant clinical strains, by generating intracellular ROS and affecting plasma membrane permeability.

Antifungal therapy of Candida biofilms: Past, present and future

Summary: Virtually all Candida species linked to clinical candidiasis are capable of forming highly resistant biofilms on different types of surfaces, which poses an additional significant threat and further complicates therapy of these infections. The scarcity and limited effectiveness of antifungal agents, particularly against biofilms, are major challenges in the treatment of Candida biofilms. However, there is optimism that these challenges can be overcome in a reasonable timeframe.

Summary: This study demonstrates, for the first time, the inhibitory effect of pseudolaric acid A (PAA) alone and in combination with fluconazole (FLC) on Candida albicans biofilms. The combination of PAA and FLC shows an improved antibiofilm effect by inhibiting adhesion and yeast-to-hypha transition. These findings provide new insights into the mechanisms of antifungal drug combinations and offer potential strategies for addressing C. albicans drug resistance.

Antifungal activity and potential mechanism of Asiatic acid alone and in combination with fluconazole against Candida albicans

Summary: This study found that Asiatic acid in combination with fluconazole had a synergistic effect on fluconazole-resistant Candida albicans both in vitro and in vivo. The combination reduced the minimum inhibitory concentration of fluconazole and inhibited hyphal growth, possibly through mechanisms such as inhibition of drug efflux pump and accumulation of reactive oxygen species. These findings provide new insights for overcoming drug resistance and offer potential for future development and application of Asiatic acid.

These findings suggest the anti-Candida effects of PAE plus FLZ/AmB and their potential to increase the sensitivity of C. albicans to FLZ/AmB.
