Peripheral immune factors aiding clinical parameter for better early recurrence prediction of hepatocellular carcinoma after thermal ablation

Fangying Fan et al.

Summary: This study aimed to explore the prognostic value of peripheral immune factors (PIFs) for early recurrence (ER) prediction of hepatocellular carcinoma (HCC) after thermal ablation. Different models were constructed and evaluated using area under the curve, recurrence-free survival analysis, and log-rank tests. The results showed that peripheral immune factors, when combined with clinical parameters, improved the accuracy of early recurrence prediction after thermal ablation for HCC, and aided in risk stratification of patients.

Summary: This real-world study found that microwave ablation (MWA) provided a promising treatment for hepatocellular carcinoma (HCC). The results showed an increasing trend in survival for HCC patients treated with MWA over time. A clear survival benefit was also observed in early recurrent patients.

NLRC3 High Expression Represents a Novel Predictor for Positive Overall Survival Correlated With CCL5 and CXCL9 in HCC Patients

Chengpan Wang et al.

Summary: NLRC3 has been reported as a factor inhibiting inflammatory responses. Its role in hepatocellular carcinoma (HCC) is still unknown. In this study, NLRC3 levels were evaluated using the GEO database and multiple immunohistochemical analysis with tumor tissue microarrays of HCC patients. Tumor-bearing mouse models were also established to assess the effect of NLRC3. The results showed that high NLRC3 expression was associated with favorable overall and disease-free survival. NLRC3 was also found to be involved in CD8(+) T cell infiltration and correlated with increased CCL5 and CXCL9 expressions in the tumor microenvironment. These findings suggest that boosting NLRC3 could be a promising treatment to enhance survival in HCC patients.

Cancer associated fibroblast-derived CCL5 promotes hepatocellular carcinoma metastasis through activating HIF1α/ZEB1 axis

Haixu Xu et al.

Summary: This study identifies a novel mechanism in which CAF-derived chemokine CCL5 enhances the metastasis and invasion of HCC through the HIF1 alpha/ZEB1 axis. This finding may have implications for the prognosis of HCC and provides a new therapeutic target for HCC.

Construction and validation of a prediction model of extrahepatic metastasis for hepatocellular carcinoma based on common clinically available data

Liuxin Zhou et al.

Summary: This study aimed to investigate the clinical characteristics and risk factors of hepatocellular carcinoma (HCC) patients with extrahepatic metastases (EHM) and establish an effective predictive nomogram. Clinical and pathological data from 607 HCC patients were analyzed and a predictive model was built using R. The model showed good predictive accuracy and discriminating ability, and was validated with a separate cohort of 151 patients. The study concluded that the constructed prediction model has some evaluation capability for HCC EHM.

Intratumoral γδ T-Cell Infiltrates, Chemokine (C-C Motif) Ligand 4/Chemokine (C-C Motif) Ligand 5 Protein Expression and Survival in Patients With Hepatocellular Carcinoma

Na Zhao et al.

Summary: The study found an increased infiltration of gamma delta T cells in the tumor microenvironment of HCC patients, positively correlated with the expression of immune-related genes. Interaction between CCL4/CCL5 and their receptors CCR1/CCR5 may facilitate the recruitment of gamma delta T cells from peripheral blood or surrounding areas to tumor regions.

Targeting metastatic cancer

Karuna Ganesh et al.

Summary: Improved understanding of metastases biology has led to advancements in targeting and treating both micro- and macrometastases. Recent studies have identified the unique biology of metastasis-initiating cells, providing new opportunities for developing more effective treatment strategies through cancer sequencing, mechanistic studies, and clinical trials. These insights offer potential for better outcomes in targeting metastatic relapse and improving patient survival.

Hepatocellular carcinoma

Josep M. Llovet et al.

Summary: Liver cancer, particularly hepatocellular carcinoma (HCC), poses a significant global health challenge with rising incidence projected to exceed 1 million cases by 2025. Infection by hepatitis B and C viruses remains a major risk factor for HCC, while non-alcoholic steatohepatitis is emerging as a more common risk factor in Western countries. Advances in systemic therapies for HCC, including immunotherapies and targeted therapies, are expected to revolutionize the management of this disease.

Zhen Wang et al.

Summary: Clinical trials have shown that Huaier granules in combination with thermal ablation can benefit HCC patients by improving overall survival, progression-free survival, and reducing extrahepatic metastatic rates. Younger patients with single tumors and tumors smaller than 3 cm may experience better outcomes with the integrated treatment approach.

Summary: This study evaluated the gadoxetate-enhanced MRI findings and prognostic significance of the proliferative class of hepatocellular carcinoma (HCC) after surgery. The proliferative class was associated with poor overall survival, increased rates of intrahepatic and extrahepatic metastasis, and specific predictive factors such as rim arterial phase hyperenhancement on MRI and high serum alpha-fetoprotein levels.

Yun-Fan Sun et al.

Summary: This study used single-cell RNA sequencing to analyze circulating tumor cells from liver cancer patients, revealing that the transcriptional dynamics of CTCs are associated with stress response, cell cycle, and immune evasion signaling. Additionally, it was identified that CCL5 plays a crucial role in immune evasion of CTCs.
