Free radicals: Relationship to Human Diseases and Potential Therapeutic applications

Finally, recent findings regarding therapeutic modulations of intracellular levels of reactive oxygen species are presented, which may have significant implications for developing novel agents for treating several diseases.

Thus, recovering phytochemicals from the waste generated during the processing of Mediterranean diet ingredients may provide a new strategy for cancer prevention and improve the economic feasibility and sustainability of the food processing industry in the region.

Glutathione-Mediated Conjugation of Anticancer Drugs: An Overview of Reaction Mechanisms and Biological Significance for Drug Detoxification and Bioactivation

Summary: The effectiveness of anticancer drugs is influenced by specific metabolites that can alter their therapeutic or toxic properties. One important process in metabolizing these drugs is conjugation with reduced glutathione, which can occur non-enzymatically or with the help of glutathione-dependent enzymes. Glutathione binds with anticancer drugs to form more polar and water-soluble glutathione S-conjugates, facilitating their excretion. This conjugation plays a role in detoxification, reducing the likelihood of the drugs reacting with cellular targets. However, some drugs become more active or toxic when transformed into thioethers through glutathione conjugation. Therefore, glutathione conjugation can also lead to pharmacological or toxicological effects through bioactivation reactions.

The tripeptide glutathione (GSH) plays a crucial role in maintaining proper redox cell signaling and limiting excessive oxidative stress. GSH activates the Keap1-Nrf2-ARE pathway to control the expression of genes that regulate antioxidants, inflammation, and the immune system. GSH depletion may be central to the pathophysiology, immune response, severity, and mortality of inflammatory diseases and COVID-19. Enhancing GSH levels could be a key therapy to reduce severity and fatal outcomes in these diseases.
