Biomarkers for Outcome in Metastatic Melanoma in First Line Treatment with Immune Checkpoint Inhibitors

Tanja Mesti et al.

Summary: A retrospective analysis showed that immune-related adverse events (irAEs) and serum immune-inflammation parameters could serve as potential biomarkers for response and survival in metastatic melanoma patients treated with immune checkpoint inhibitors. Patients with high pre-treatment levels of systemic immune-inflammation index (SII) had a higher risk of progression and death, but those with irAEs in the high-SII group might respond well to immune checkpoint inhibitors.

Modeling the effect of gut microbiome on therapeutic efficacy of immune checkpoint inhibitors against cancer

Yu Chen et al.

Summary: Modulating the gut microbiome can enhance the efficacy of immune checkpoint inhibitors. The levels of favorable and unfavorable gut bacteria can predict patients' response to anti-PD-1 therapy. Non-responders' gut microbiome composition can be manipulated to improve response rates and achieve long-term progression-free survival.

Interferon-γ: teammate or opponent in the tumour microenvironment?

Angela M. Gocher et al.

Summary: IFN gamma plays pleiotropic roles in the tumor microenvironment, acting as both a 'teammate' to promote antitumor immune responses and an 'opponent' promoting tumor growth and suppressing immune responses.

A review on the role of gut microbiota in immune checkpoint blockade therapy for cancer

Esther Kim et al.

Summary: The gut microbiome and gut cytokines have shown potential as biomarkers for carcinogenesis and predicting response to immune checkpoint inhibitors, playing a significant role in the effectiveness of PD-1 and CTLA-4 blockade. Understanding the interplay between microbiome, microbiome-generated cytokines, and immune checkpoints may lead to new insights in immunotherapy.

Changes in expression of PD-L1 on peripheral T cells in patients with melanoma and lung cancer treated with PD-1 inhibitors

Sarah J. Dart et al.

Summary: This study found that patients with a higher proportion of PD-L1(+) T cells at baseline had improved objective response to PD-1 inhibitor therapy, while patients with a lower proportion of regulatory T cells at baseline experienced more immune-related adverse events. These findings may assist in clinical decision making.

Prognostic significance of programmed cell death-ligand 1 expression on circulating tumor cells in various cancers: A systematic review and meta-analysis

Yushu Ouyang et al.

Summary: Our meta-analysis found that PD-L1 positive expression on CTCs predicted better survival prognosis for ICI treatment but worse survival for other therapies. Post-treatment PD-L1(+) CTCs predicted worse PFS and OS. The prognostic value of PD-L1(+) CTCs was significantly modulated by comparison models.

Adverse events during immunotherapy in Slovenian patients with metastatic melanoma reveal a positive correlation with better treatment outcomes

Tanja Mesti et al.

Summary: Patients with metastatic melanoma treated with immunotherapy who developed immune-related adverse events showed better treatment outcomes, with longer progression-free survival and higher overall response rates, compared to patients without immune-related adverse events.
